【Objective】To study the antifungal activity of compounds from HuangTeng.In order to find efficient,cheap antifungal agents with less side effect.【Methods】antifungal activity of chloroform and methanol exteacts from HuangTeng plant against Candida albican,Candida glabratas,Candida parapsilosis, Candida krusei and Cryptococcus neoformans was studied by hole agar diffusion test, the antifungal activity was evaluated by diameter of antibacterial circle.The MIC was determined in accordance with the National Committee for Clinical Laboratory Standards(NCCLS)guidelines J(M27-A).We investigated the combination effect of fluconazole(FLC)and compounds on FLC-resistant Candida albican by checkerboard microdilution assay,and the effect was evaluated by calculating the fractional inhibited concentration index(FICI).Agar disk diffusion test,time-kill curve studies confirmed the synergism of them.【Results】The root,bark,core and rattan parts of HuangTeng have similar antifungal activity,the antifungal activity of chloroform and methanol exteact is similar to each other,diameter of antibacterial circle was 10~20mm.Chemical compounds from HuangTeng have stroger antifungal activity,average MIC was 80~640μg/ml.8-Hydroxy Roemenine which has antifungal activity is newly-found chemical compound from HuangTeng,MIC was 320~640μg/ml.When compounds were tested by being combined with fluconazole,both compounds and fluconazole displayed significant activity with lower MIC against FLC-resistant Candida albican, synergic antifungal effect was observed when they were used in combination (FICI<0.5).Agar disk diffusion test displayed that antifungal effect was poor when drug was used alone,however,antifungal effect become stroger when two drugs were in combination.Time-kill curve studies also confirmed the synergic effect.【Conclusion】Compounds from HuangTeng have antifungal activity,it can reverse Candida albican's resistance to fluconazole.
