Objective To establish the experimental model of distraction osteogenesis and bone transportfor the treatment of bone defect in rabbits. To evaluate the effects of osteogenesis in bone transportfor the treatment of large segmental bone defect by using an injectable tissue engineered bone withplatelet-rich plasma.Methods Thirty rabbits were divided into three groups. Twenty rabbits were created a 20mmincision in the distal tibia shaft in addition to the metaphysis osteotomy,then stabilized with anself-designed mini external fixator bilaterally. From post-operative day 7 onwards,bone fragmentswere distracted by 1 mm a day. After elongation, percutaneous injection of injectable tissueengineered bone in the distracted gap of experimental group. X-ray analysis, histological andradionuclide imaging examinations were performed at postoperative weeks (POW)2,6 and10. And bone mineral absorptiomely analysis were carried out at POW 2,4 and 8 .Results Radiographically and Radionuclide imaging.the experimental group,the bone formingactivity,bone healing speed and remodeling were all significantly better than those in controlgroup. Radionuclide imaging statistical analysis of vatiance of the repeated measures datademonstrated significant difference by injection of injectable tissue engineered bone in thedistracted gap. The differences of BMD between two groups were statistically signifificant( P < 0.05).Conclusion Percutaneous injection of injectable tissue engineered bone with Platelet-richplasma in bone transport for the treatment of large segmental bone defect is beneificial forregeneration,mineralization and remodeling. It can shorten the consolidatin period.
