Objective: To study and analysis the mechanism of action and inhibitory effect of a new type paclitaxel nanoparticle (TAX-NLC) on KB and SKOV3 cells for developing a new targeted pharmaceutical to human tumor.Methods: F-actin,Ca2+ and Apoptosis dyed by Tunel of KB and SKOV3 cells after being exposed to TAX-NLC were observed by laser confocal microscopy. Nuclear dyed by Dapi and Giemsa of KB and SKOV3 cells after being exposed to TAX-NLC was observed by fluorescence microscope and light microscope. Cultivate KB and SKOV3 cells with TAX-NLC for 3 hours respectively and then cultivate 12,24,48,72 hours without TAX-NLC , utilize MTT method to see its inhibitory effect . Count the number of KB and SKOV3 cells after being treated with TAX-NLC or TAX for 3 hours and then cultivate 1,2,3,4,5,6 days, using trypan blue refused dyeing method .Results: After tumor cells were exposed to TAX-NLC, F-actin of the two cells transformed, from regular filament to disorder clumps or spot distributinon, and the cystoskeleton collapsed. Compared to control group, intracellular Ca2+ of treatment group increased obviously, and about 1 hours later reach the peak. TAX-NLC can induce apoptosis, Apoptotic index of SKOV3 cells induced by TAX-NLC was higher than KB cells. What's more, we find polykaryocytes in both KB and SKOV3 cells after exposed to TAX-NLC. The inhibition ratio of TAX-NLC to KB or SKOV3 cells was increased with the increasing of dose and time, and the highest inhibition ratio was 92% . By cell growth curve, the inhibitory effects of TAX-NLC and TAX on SKOV3 and KB cells were obviously, however.Conclusions:TAX-NLC actively takes effect on F-actin and intracellular Ca2+ ,and induce apoptosis in KB and SKOV3 cells. Also, it can obviously inhibit the growth of tumor cells. The new developed TAX-NLC maybe a new potential targeting therapeutic system to human tumor.
