Study Of Molecular Mechanism Of Imbalance Of Immune Homeostasis And Chinese Medicine Intervention Of Kidney Deficiency Constitution Rat

Objective: To improve construction methods of kidney deficient constitution rat model, in-depth study of constitution characteristics of kidney deficient constitution, through detection of the immune cytokine and T-lymphocyte expression,discuss molecular mechanism of immune homeostasis imbalance of kidney deficient constitution rat.By drugs of improvement kidney and spleen function intervention,Observation adjustment effect of drug contrapose immune homeostasis imbalance of kidney deficient constitution rat,provide experimental basis for the character of kidney deficient constitution and correlative disease awareness and prevention.Methods: Based on the well-known argument of chinese medicine " intimidation damnification kidney ", adopt "cat scared rats" classical modeling method, Result Shenjing damnification of maternal rats ,and mice of offspring congenitally deficiency animal models of kidney deficient constitution。By drugs of improvement kidney and spleen function intervention pregnant maternal rats, observe of their offspring the formation of kidney deficiency, detection immune indicators of offspring mouse IL-10, IFN-γand T lymphocytes CD4+, CD8+ molecule expression, do statistical analysis.Results: The offspring mouse serum IL-10, IFN-γcontent changes in each group is obvious. Test results of serum IL-10 showed that:IL-10 levels of intimidation model group, drug comparison group have increased (P <0.05). Test results of IFN-r shows that:IFN-r levels of intimidation model group were significantly lower (P <0.01), IFN-r of drug control group were decrease (P <0.05).Test results of CD4+ and CD8+ showed that:CD4+, CD8+ percentages of kidney deficiency constitution offspring rat spleen lymphocytes suspension were lower than the natural group (P <0.05).Conclusion: Under the guidance of the viscera theory of Chinese medicine, use method of threat to making kidney damnification to manufacturing model of kidney deficiency constitution having theoretical and practical feasibility,kidney deficiency constitution rat is a marked imbalance of immune homeostasis. Through intervention of improvement kidney function drugs can improve the imbalance in rat kidney deficiency of the immune homeostasis, drug intervention group of rats tended to a number of indicators are being normal.
