Objective: To study the expression of transcription factor T-bet of T helper 1 type cells in peripheral blood mononuclear cells(PBMNC) in aplastic anemia mice and the effect of cyclosporine A(CsA)on T-bet in PBMNC in normal mice stimulated by mice bone marrow macrophage cell lines Ana-1.Methods:①Immunologically mediated method was used to establish the model of aplastic anemia in BALB/c mice.②T he expression of T-bet mRNA in PBMNC in aplastic mice was measured by reverse transcriptase polymerase chain reaction(RT-PCR).③PBMNC in normal mice were stimulated by supernatant of cultured myeloid matrix cells in aplastic mice and mice bone marrow macrophage cell lines Ana-1 and supernatant of cultured Ana-1 cell lines adding to CsA respectively, and the expression of T-bet mRNA was measured by RT-PCR.Results:①T he positive rate of T-bet of AA mice was 17/20.In normal controls,it was 3/15.Moreover the expression intensity of T-bet was higher in AA mice than the normal controls.②Stimulated by supernatant of cultured myeloid matrix cells in aplastic mice and Ana-1 cell lines, the positive rates of T-bet in PBMNC in normal mice were 12/15 and 14/15. In normal controls,it was 3/15.And the expression intensities of T-bet were 1.29±0.03 and 1.54±0.03 respectively,higher than that of normal controls (1.03±0.06) (P<0.05).③Stimulated by supernatant of cultured Ana-1 cell lines adding to CsA, the positive rate of T-bet in PBMNC in normal mice was 2/15,while only stimulated by supernatant of cultured Ana-1 cell lines,it was 14/15.The former expression intensity of T-bet was 0.66±0.02,less than the latter(1.54±0.03)(P<0.05).Conclusion: The results indicate macrophage may participate in immune pathogenesis of aplastic anemia through increasing the expression of T-bet and it may be one of its immunosuppressive effects in treatment of AA that CsA restrains the produce of T-bet.
