Purpose:Due to immune-mediated aplastic anemia mouse model to study the object, and Observe the effective of Yisui Shengxue granules on abnormal apoptosis of CD34~+ cells in aplastic anemia mice due to immune-mediated. This study aims to explore the effective mechanisms of the granules on the aplastic anemia and explain the theory of"the Kidney governs bones to generate marrows"with modern biological theories and methods.Material and method:In this study, 48 SPF level BALB / c mice, were randomly divided into three groups, namely, the normal group, the aplastic anemia model group (model group) and the Yisui Shengxue granules treatment group (treatment group). The aplastic anemia model in mice was built with immune-mediated method. The apoptosis of CD34~+ cells in bone marrow was detected with flow cytometry method,and observe the benefits of blood marrow particle mechanism for the effective mechanisms of the granules on the model was observed.Results:The model group and treatment group mice peripheral blood as red blood cells, white blood cells, hemoglobin, platelet, reticulocyte in peripheral blood of the mice significantly decreased in model group and treatment group. And percentage of CD34~+ cells apoptosis in bone marrow of the mice significantly increased in the groups. But the lower level changes of peripheral blood and the upper level changes of percentage of CD34~+ cells apoptosis in the treatment group were significantly more less than that of the model group.Conclusion:Yisui Shengxue granules can treat aplastic anemia mice induced by immune-mediated in a certain degree. Yisui Shengxue granules can increase the peripheral blood level and decrease percentage of CD34~+ cells apoptosis of the mice.It is revealed that Yisui Shengxue granules can improve bone marrow function recovery by affecting the changes of peripheral blood and percentage of CD34~+ cells apoptosis to the immune-mediated aplastic anemia mice.This study results provide a modern scientific evidence for the theory of"the kidney governs bones to generate marrow"in TCM. |
