Anti-Bladder Tumor Effect Of Single Using Ag85A DNA Vaccine And Ag85B DNA Vaccine On Bladder Tumor Rat

Objective: To explore the immunological effect of Single Using Ag85A DNA vaccine and Ag85B DNA vaccine on rat with bladder tumor.Methord: First Using the female Wistar rat, Carcinogen MNU irrigated bladder in order to construct the model of bladder tumor. We randomly divided 40 rats into the five groups: NS, pcDNA3.1, BCG, Ag85A DNA vaccine and Ag85B DNA vaccine, 8 rats in each group. Immuned time is the seventh day, fourteenth day and twenty-first day after model succession. At the twenty-eighth day, the rats were sacrificed. Spleen was removed aseptically. The percentages of CD4+T cell and CD8+T cell were measured by performing flow cytometry. And IFN-γin sera of the bladder tumor rats were assayed with ELISA. In addition, Bladder tumor was did pathology detection.Results:1. pcDNA-Ag85A plasmids and pcDNA-Ag85B plasmids were extracted from E.coli JM109.2. The model of bladder tumor on rat was constructed successfully.3. After the DNA vaccine treatment, the bladder tumor size in Ag85A DNA vaccine group and Ag85B DNA vaccine group became smaller, the tumor grade became better.4. The result of the percentage of CD4+T cell, CD8+T cell and the relative value of CD4+/ CD8+ showed that there was significant difference between the Ag85A DNA vaccine group, Ag85B DNA vaccine group and the control group (P<0.05), and Ag85B DNA vaccine is stronger than Ag85A DNA vaccine (P<0.05), But the effects still failed to come up to the level of BCG. (P<0.01).5. Immunization of rats with Ag85A DNA vaccine and Ag85B DNA vaccine could increase the production of IFN-γ,and that of Ag85B DNA vaccine is more than Ag85A DNA vaccine ( P<0.01), But still less than BCG.(P<0.01).Conclusion:1. The Ag85A DNA vaccine or The Ag85B DNA vaccine could induce strong immune response of bladder tumor rat. But its effect still fail to come up to the level of BCG..2. Anti-Bladder Tumor effect of Ag85B DNA vaccine is stronger than Ag85A DNA vaccine.
