Objective:To discuss the relationship of cell apoptosis and proliferation in different differentiation degree of chondrosarcoma by checking in its variation regularity in generation, development, differentiation and prognosis of chondrosarcoma.Methods:1 Cases: all sample were from the second hospital of Shanxi Medical University. Experienced pathological doctors examined HE stain slice again to determine pathological diagnosis.2 Experimental group: 21 cases chondrosarcoma. By Enneking surgical staging, stage I 11 cases, stage IIa 6 cases, stage IIb 3 cases, stage III 1 cases. WHO Tumor cell differentiation: High differentiation 11 cases, moderate differentiation 5 cases, poor differentiation 5 cases. Control group: 5 cases enchondroma and 5 cases osteochondroma.3 Cell apoptosis state was detected by TdT-mediated DNA in situ end labeling(TUNEL) method. Calculate apoptosis index(AI) and analyze the relationship with proliferation index(PI).4 The expression of Ki67 and S-100 and Vimentin were detected by immuno- histochemical ElivisionTM method in chondrosarcoma and enchondroma and osteochondroma.Results :1 In high and moderate and poor differentiation chondrosarcoma the apoptosis index(AI) was significantly higher than enchondroma and osteochondroma(P<0.0001).2 In moderate and poor differentiation chondrosarcoma the proliferation index(PI) was significantly higher than high differentiation chondrosarcoma and enchondroma and osteochondroma(P<0.0001).3 There was not a statistical significance in values of AI and PI between different differentiation degree of chondrosarcoma.4 The positive rates of Vimentin in all cases was 100%. The positive rates of S-100 protein in high differentiation, moderate differentiation of chondrosarcoma and enchondroma and osteochondroma was 83.9%, the negative rates of S-100 protein in poor chondrosarcoma was 16.1%.Conclusion:1 The apoptosis index(AI) of moderate and poor differentiation is significantly higher than enchondroma and osteochondroma, but it is no difference between different differentiation degree of chondrosarcoma.2 The proliferation index(PI) is significantly higher between moderate and poor differentiation chondrosarcoma with high differentiation chondrosarcoma, enchondroma and osteochondroma.3 It will be helpful to evaluate the development, progression and prognosis of chondrosarcoma by both PI and AI detection.
