Objective:(1)To observe expression of P-selectin on vascular epithelium in smoked rat penis.(2)To observe morphology of penis cavernous tissueMethod:(1)50 healthy Wistar rats were divided into 5 groups,normal group,long-time and big-dose smoking group,long-time and small-dose smoking group,short-time and big-dose smoking group,giving-up smoking group.Erectile function was tested by injecting apomorphine(APO).(2)The expression of p-selectin on vascular epithelium in smoked rat penis were measured by ELISA.(3)To observe morphology of penis cavernous tissue with transmission electron microscope(TEM).(4)Statistical analysis:Values were given as means±SE and analysized by SPSS11.0 software.Differences were considered significant at a value of P<0.05.Results:(1)The expression of P-selectin in normal group was 10.78±1.71ng/L;the expression of P-selectin in long-time and big-dose smoking group was 62.62±5.95 ng/L; the expression of P-selectin in long-time and small-dose smoking group was 40.06±3.97 ng/L; the expression of P-selectin in short-time and big-dose smoking group was 41.37±4.06 ng/L; the expression of P-selectin giving-up smoking group was 22.80±3.15 ng/L. Compared with control group,The significant diferent expression is observed in other groups(P<0.05).(2)Abnormal arrangement of endothelial,penile cavernous sinus and smooth muscle cell were detected by electron microscope. Disruption of continuity and integrity of endothelial cell were also found.Significant changes of ultrastructure were observed in the endothelial and smooth muscle cell penile cavernous tissue in smoked rats。The proliferation of mesenchymal tissue were also observed. Conclusion:The erectile function of penis is affected seriously by smoking,and these pathological changes of ultrastructure in penile cavernous tissue may be one of the most important mechanisms of decreased erectile function in smoked rats.
