| Objective To investigate the relationship between single nucleotide polymorphism(SNP) at position 276 in adiponectin gene and type 2 diabetic nephropathy in Hans ethnicity in Shanxi province in China.Methods Seventy-six non-diabetic controls,108 type 2 diabetic patients without diabetic nephropathy(DN) and 78 type 2 diabetic patients with DN were genotyped for SNP276 of adiponectin gene by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).Their clinical parameters were also examined.Results(1) There was difference in the genotypes and allele frequency distributions of SNP276 between type 2 diabetes mellitus(T2DM) patients and controls(P<0.05).The frequencies of G allele in T2DM group were higher than that in normal control group (P<0.01).The risk of T2DM increased in GG or GT genotype compared to TT genotype.In the distribution of genotypes and allele frequencies of the SNP276 polymorphism,there was no statistical difference between T2DM with DN and T2DM without DN(P>0.05).(2) The body mass index(BMI),systolic blood pressure,fasting plasma insulin,HOMA-IR and fasting plasma glucose were higher in GG or GT genotype than in TT genotype in control subjects.The systolic blood pressure,fasting plasma insulin HOMA-IR and fasting plasma glucose were higher in GG or GT genotype than in TT genotype in T2DM without DN group.The systolic blood pressure,HOMA-IR was higher in GG or GT genotype than in TT genotype in T2DM with DN group.Conclusion Adiponectin gene SNP276 polymorphism associated with type 2 diabetes in Shanxi Han population,but IT was not associated with DN.The G allele may be a risk factor of insulin resistance and type 2 diabetes in Han ethnicity of China. |
