Objective To detect the serum protein markers in patients with lymphoma by the surface enhanced laser desorption/ionization-time of flight-mass spectrometry(SELDI-TOF-MS) protein chip technique,and to screen biomarker candidates and identify serum specific protein biomarkers in order to study diagnostic value.Methods The SELDI-TOF-MS technique and weak cation exchanger(WCX2) protein chip were used to detect the serum differentially expressed protein markers of 96 patients with lymphoma and 100 healthy volunteers.The sensitivity and specificity of lymphoma protein mass spectrum to diagnose lymphoma were screened.The sensitivity of lymphoma protein mass spectrum were compared between stageⅠ-Ⅱand stageⅢ-Ⅳ,so were in group A and group B.Furthermore,the sensitivity of LDH,β2-MG,CA125 of lymphoma tumor markers and protein mass spectrum were compared.Results(1) There were fifteen specific marker proteins which have a significant difference (P<0.05) between lymphoma patients and healthy volunteers.Among the total,the protein markers of 2047Da,2819Da,2867Da,3375Da,5810Da,7974Da and 8078Da were high level in patients.Correspondingly,the protein markers of 3398Da,3973 Da,6436Da,6629Da,6880Da,8564Da,8692Da and 13751Da were low level in patients.(2) Four protein peaks(6880Da,8564Da,8692Da and 13751Da) were low level in lymphoma patients,and they could be used to distinguish lymphoma patients from healthy people.Their sensitivity and specificity were 82.29%and 80.00%,78.13%and 73.00%, 75.00%and 80.00%,71.58%and 83.00%in lylnphoma diagnosis,respectively.(3) When they were used to diagnose lymphoma in stageⅠ-Ⅱ,the sensitivity of four protein peaks were 77.55%,71.43%,71.43%and 67.35%,respectively.When they were used to diagnose lymphoma in stageⅢ-Ⅳ,the sensitivity of four protein peaks were 87.23%, 85.11%,78.72%,76.6%,respectively.(4) When lymphoma patients did not have general symptom(in group A),the sensitivity of four protein peaks were74.58%,72.88%,69.49%and 64.41%,respectively.When lymphoma patients had general symptom(in group B),the sensitivity of four protein peaks were 86.49%,86.49%,86.49%and 78.38%,respectively.(5) The sensitivity of four protein peaks(6880Da,8564Da,8692Da and 13751Da) were higher thanLDH,β2-MG,CA125,respectively(P<0.05).Conclusion Protein mass spectrums of 6880Da,8564Da,8692Da and 13751Da which have been detected by the SELDI protein chip technique are hoped to be serum specific biomarkers for lymphoma diagnosis.
