Chinese herbs, which is made up of plants, animals and minerals, are natural resources, which can be absorbed most easily by animals. Furthermore Chinese herbs, which have little side-effect, little remains, little drug-resistance and lead to little pollutions, can bring fine effects on dairy cows. Thus, people use Chinese herbs as raw materials to product mixed additives without antibiotics. Research and use of additives on milk cows are the focus of current scholarly pursuit. Such Chinese traditional medicines as liquorice, Semen Vaccariae, and etc contain a lot of nutritious substance and biologically active substances. They can supply organism with nutrition, improve creatural appetite, digesting and assimilating, metabolism, blood circulating of organism. They also can kill microorganism, improve immunity of organism, prevent and cure diseases, accordingly improve lactation capability of milk cows.The study, as following, was on the level of biochemistry and molecular biology. We selected five Chinese herbs among the compound recipes extensively used in human beings to increase the level of milk, which were Cowherb Seed, JiaZhu, Rhaponticum uniflorum, Medulla Tetrapanacis and Taraxacu Mogono. Juice from the herbs after boiling was extracted. The cells of mammary gland of mice that fed their babies more than 5 days and less than 12 days were used. The herbs were added into the culture medium of cells respectively and series of comparisons were done. the expression ofβ-Casein compared with the circumstances without additive and only within prolactin. There were five parts in the experiment. First, we used the approach of MTT to evaluate how the herbs influenced the cells on cell proliferation. Second, we studied the regulation of cyclin D1 and Stat 5 by laser scanning Confocal influorescence microscopy. Third, Real time Quantitative PCR was used to detect the expression of mRNA ofβ-Casein. Fourth, capillary electrophoresis was used to detec the content ofβ-Casein. Fifth, different juices were injected into the mammary glands of mice of lactating on lactation to measure quantities of milk respectively .The result showed that pangolin scales, medulla tetrapanacis and rhapontici could raise the level of multiplication of mouse mammary gland epithelial cells(P<0.05). Dandelion can raise the level of multiplication of mouse mammary epithelial cells extremely(P<0.01). Cowherb seed has no obvious influence on mouse mammary gland epithelial cells. Cowherb seed can raise the level of cyclinD1 extremely, while pangolin scales, medulla tetrapanacis and dandelion can raise the level of cyclinD1 significantly(P<0.05), and radix rhapontici has no obvious effect on the level of cyclinD1. And, medulla tetrapanacis, dandelion and radix rhapontici can significantly promote the expression of beta-casein, while cowherb seed and pangolin scales both can significantly promote the expression of beta-casein extremely. There were some effects in bothβ-Casein expression and cell proliferation that were identified partly by the study.The significance of the research, on the level of biochemistry and molecular biology, extended the research on the mechanism of action of herbs .
