Backgroud:Preeclampsia is the main cause of the increasingly higher case fatality and mortality of pregnant wo号men and infant.However,the mechanism of preclampsia are still unclear.Recent data have revealed that Preeclampsia is intensively related with the Immunity Imbalance,which included changing in cytokine.Suppressor of cytokine signaling(SOCS) is a new family of immuno-suppressor,including CIS,SOCS1-SOCS7 and so on. The functions of SOCS have been revealed more and more since its discovery in 1995,such as negatively regulating signaling during the embryonic stage of development and so on.SOCS proteins have emerged as critical regulators of cytokine-mediated signaling in diverse tissues.These proteins act in a negative feedback loop to attenuate signaling via the Janus kinase-Signal transducers and activators of transcription(JAK/STAT) pathway.SOCS3 is one of the most active member of SOCS family.SOCS3 is normally activated by JAK/STAT signaling in normal cells,which in turn inhibits JAK activity and subsequent STAT3 activation.Decreased SOCS3 activity in human gestational tissues may play a part in the onset/progression of term labor.Since SOCS proteins negatively regulate cytokine-mediated inflammatory processes.As SOCS3 regulated the differentiation of Th cells,inhibiting Th1 cells and stimulating Th2 cells. Also,there are immunity imbalance of Th1/Th2 cytokines in the Preeclampsia. So,it's presumed that SOCS3 ingage in the pathological reason of preeclampsia.Objective:To investigate the mRNA and protein expression and location of SOCS3 in Preeclampsia amnion,choriodecidua and placenta.Methods:1.SOCS3 mRNAs were detectable by RealtimaPCR in preeclampsia amnion, choriodecidua and placenta.2.SOCS3 proteins were detectable by western immunoblotting,in preeclampsia amnion,choriodecidua and placenta.3.Immunohistochemistry analysis revealed the location of SOCS3 in preeclampsia amnion,choriodecidua and placenta.Results:1.The mRNA transcripts for SOCS3 were detectable by RealtimePCR in preeclampsia human amnion,choriodecidua and placenta,and the positive rate of SOCS3 was significantly lower in preeclampsia group than in normal group(60.0%,20.0%,26.7%);2.SOCS proteins were,in general,significantly more abundant in placenta compared with amnion or choriodecidua.3.SOCS3 protein expression were in consistent with mRNA overall.4.SOCS3 immunoreactive protein was detected in syncytial and mesenchymal in preeclampsia group.Conclusions:1.The relatively large amounts of SOCS proteins in the placenta may reflect a placenta-specific immunoprotective response to minimize the elaboration and effects of cytokines with potential to harm the placenta and fetus.2.SOCS3 mainly exert roles in syncytial and mesenchymal.3.The decrease of SOCS3 are closely correlated with the Preeclampsia.Detecting these indexes can be helpful in assessment of the biological behaviors of Preeclampsia.
