Objective:To investigate the expression and with the relationship of clinical pathology of estradiol(E2),testosterone(T),estrogen receptor(ER),androgen receptor(AR),17β-hydroxysteroid dehydrogenase2(17β-HSD2) and 17β-hydroxysteroid dehydrogenase5(17β-HSD5) in gastric carcinomas.Methods:1,To detect the protein expression of estradiol(E2),testosterone(T),estrogen receptor(ER) and androgen receptor(AR) in samples of 56 cases of gastric carcinoma and 56 cases of normal gastric tissue by tissue microarray technology and immunohistochemical Elivision plus method.2,To detect mRNA level of 17β-hydroxysteroid dehydrogenase2(17β-HSD2) and 17β-hydroxysteroid dehydrogenase5(17β-HSD5) in samples of 56 cases of gastric carcinoma and 56 cases of normal gastric tissue by RT-PCR method.Results:1. Immunohistochemical Elivision plus method:(1). The E2 protein expression level was 0.2381±0.0718 in gastric carcinoma tissues,whileas that was 0.1832±0.0628 in normal gastric tissues (P<0.01).(2). The T protein expression level was 0.1985±0.0636 in gastric carcinoma tissues,whileas that was 0.1946±0.0659 in normal gastric tissues (P>0.05).(3). The ER protein expression level was 0.1729±0.0361 in gastric carcinoma tissues,whileas that was 0.1666±0.0373 in normal gastric tissues (P>0.05).(4). The AR protein expression level was 0.1771±0.0588 in gastric carcinoma tissues,whileas that was 0.1774±0.0486 in normal gastric tissues (P>0.05).(5). Compared with the group of no lymph node metastasis, the E2 protein expression level of lymph node metastasis group was obviously higher(P<0.01).The E2 protein expression level was positively related with clinical stages. And the level of E2 protein had ascending trends according to the clinical stages. But there was no significant difference between E2 protein and other clinicopathological features such as sex, age, tumor position and differentiation status(P>0.05).2. RT-PCR method:(1). The 17β-HSD2 mRNA lever was 0.8065±0.3797 in gastric carcinoma tissues,whileas that was 0.9788±0.2325 in normal gastric tissues (P<0.05).(2). The 17β-HSD5 mRNA level was 0.5536±0.4729 in gastric carcinoma tissues,whileas that was 0.6332±0.4488 in normal gastric tissues (P>0.05).(3). Compared with the group of female, the 17β-HSD2 mRNA lever of male group was obviously higher(P<0.05).The 17β-HSD2 mRNA lever was positively related with differentiation status. And the level of 17β-HSD2 mRNA had ascending trends according to the differentiation status. But there was no significant difference between 17β-HSD2 mRNA and other clinicopathological features such as age, tumor position,lymph node metastasis and clinical stages(P>0.05).3. Expression of E2 was closely negatively correlated with that of 17β-HSD2 in human gastric carcinomas (r=0.294,P<0.05).Conclusions:1. Expression of E2 protein are highly up-regulated in gastric carcinoma.2. Expression of 17β-HSD2 mRNA are highly down-regulated in gastric carcinoma.3. The abnormal expression of E2 and 17β-HSD2 may play important roles in the tumorigenesis and progression of gastric and have the important significance in endocrine therapy and the study of selective 17β-HSD moderator.
