| Objective:we evaluate the change of estrogen metabolic enzymes before and after the treatment, and study the distinction between the different groups and between ectopia and positional endometrium, in order to obtain the possible target point of the therapy of expelling dampness and dissipating blood stasis in the treatment of adenomysis.Methods:we firstly established animal model of adenomysis by the method of transplating the pituitary gland of male mice into the uterus of female mices, then we devided the adenomysis mice into seven groups (12 mices for each group):high dose of Neiyikangfupian (NYKFP) group, medium dose of NYKFP group, low dose of NYKFP group; Danefukangjiangao(DEFKJG) group; Gestrinone group; 3 months group and 6 months group. We also had sham-operation group and blank group. Feeding by physiological saline for 3 months, we firstly executed the sham-operation group, blank group and 3 months group during the oestrus or anoestrus. While other groups healed by different medicine of group names, and 6 months group still fed by physiological saline for another 3 months, then we executed all groups during the oestrus or anoestrus. We observed and analysed the endometrum glands and stroma invasion degree of muscular layer by pathological section and HE stain, and we surveyed the change of 17β-HSDâ… ã€17β-HSDâ…¡ by the technique of immunohistochemisty.Results:1.We observe ectopic tissues on muscular infiltration and evaluate the degree of adenomyosis severity, normal group and sham-operation group is 0, the oestrus of 3 months model group is 2.67, while anoestrus is 2.17, the oestrus of 6 months model group is 3.50, while anoestrus is 2.83, the degree indicates that 3 months group endometrial stromal cells and glands infiltrate muscles layer, 6 months group endometrial stromal cells and glands infiltrate inside and outside the muscle layer, which shows that both groups molding success, and disease severity increases with the course of the disease, operation process of building had no effect.2.17β-HSD â… ,17β-HSD â…¡:in the uterus endomembrane, the the value of 17β-HSD â… and 17β-HSD â…¡ gradually increase after the construction of mice model of AM,6 months group is higher than 3 months group (P<0.05),6 months group and 3 months group are notably higher than normal group and sham-operation group (P<0.01),the compare of normal group and sham-operation group has no statistic signification (P>0.05). Compared with 6 months group, the indicates are all regulated down after treatment, NYKFP high dose group, DEFKJG group and gestrinone group has statistic significance (P<0.05)Conclusions:1. We observe ectopic tissues on muscular infiltration and evaluate the degree score of adenomyosis severity, the degree shows that both groups molding success, and disease severity increases with the course of the disease, operation process of building had no effect. The molding method of male rat pituitary transfer in female rat Uterine cavity, which molding 6 months the success rate is 100%(12/12), it is a worthy of promotion AM molding method.2. The therapy of expelling dampness and dissipating blood stasis can regulate down the estrogen metabolic enzymes expression, restrain estrogen production to treat AM. So we can say 17β-HSD â… , 17β-HSD â…¡ are the therapeutic target spots.3. NYKFP high dose group has a signification influence to the estrogen metabolic enzymes 17β-HSD â… ,17β-HSD â…¡, which is same as the Danefukangjiangao(DEFKJG) group and Gestrinone group。4.17β-HSDI has a difference between eutopic and ectopic endometrium. The prescription which have the effect of expelling dampness and dissipating blood stasis may regulate the expression of estrogen metabolic enzymes on eutopic and ectopic endometrium separately. It needs further experimental studies to confirm the regulatory mechanisms. |
