Association Between β3-AR T190C, MTHFR C677T Genes Polymorphism And Cardiovascular Risk Factor And The First Of Cardiovascular And Cerebrovascular Events In Hypertensive Patients

Objective: To investigate the association between polymorphisms ofβ3 adrenergic receptor(β3-AR) and N5,10-methylene tetrahydrofolate reductase(MTHFR) with cardiovascular risk factor and the first of cardiovascular and cerebrovascular events in hypertensive patients.Methods: 372 essential hypertention(EH) and 306 normal tensive(NT) were collected, and then DNA was extracted from their peripheral blood.With the DNA we examined theirβ3-AR T190C and MTHFR C677T polymorphisms using PCR and RFLP.Basic clinical data such as age,sex,BMI,creatinine,uric acid,triglyceride,fasting plasma glucose, course of hypertension and the first of cardiovascular and cerebrovascular events in hypertensive patients was collected, at last all the datas were analyzed by analysis of Variance,Covariance,Chi-square Test,Logistic Regression and Kaplan- Meier.Result: 1.The frequencies of different genotypes ofβ3-AR gene and MTHFR gene were in agreement with Hardy-Weinberg equilibrium in our study(P>0.05). 2.β3-AR gene showed no significant difference in genotype frequencies distribution in the two groups(P>0.05).3.MTHFR gene showed a significant difference in genotype frequencies distribution in the two groups(P<0.05), and after partition of chi-square CT+TT genotype frequencies in EH group were significantly higher than those in normal tensive (P<0.05).4.The BMI and triglyceride level ofβ3-AR gene TT, TC and CC genotype were differences,and the BMI and triglyceride level ofβ3-AR gene CC genotype were significantly higher than that of TT after analysis of Covariance(P<0.05).5. The serum uric acid level of MTHFR gene CC,CT and TT genotype were differences,and after multiple comparisons analysis the serum uric acid level of MTHFR gene TT genotype were significantly higher than that of CC (P<0.05).6.Logistic regression showed that after adjusting other confounding factors, MTHFR gene CT+TT genotype were significantly associated with EH, the odds ratios were 2.827. Age, BMI and MTHFR gene CT+TT genotype were independent risk factors associated with EH after logistic regression analysis.7.Hypertensive Patients withβ3-AR gene TC+CC genotype maybe easier to happen the first of cardiovascular and cerebrovascular events than TT genotype(P<0.05).But hypertensive patients with MTHFR gene CT+TT genotype cannot increase the risk of the first of cardiovascular and cerebrovascular events(P>0.05).Conclusion:β3-AR gene polymorphisms was significantly associated with triglyceride level and BMI,but not with Uric acid level and EH;however MTHFR gene polymorphisms was significantly associated with Uric acid level and EH,but not with triglyceride level and BMI.Hypertensive Patients withβ3-AR gene TC+CC genotype maybe easier to happen the first of cardiovascular and cerebrovascular events than TT genotype.But hpertensive patients with MTHFR gene CT+TT genotype cannot increase the risk of the first of cardiovascular and cerebrovascular events.
