[Objective]1.To explore the distribution of bile duct collagen fiber and the different species difference between animals.2.To discuss variety of ER and PCNA expression of cholangiocytes of hepatohilar bile duct with bile duct ligation , and to explore relationship between estrogen and cholangiocytes proliferation of hepatohilar bile duct.[Methods]1.The changes under stress effect and collagen fiber distribution of vertical and horizontal cross-section of human and rat bile duct were observed through normal and expanding hepatohilar bile duct collagen fiber Masson trichrome staining in human and rat.2.SD rats were randomized into the control group, bile duct ligation(BDL) group and BDL treated with tamoxifen group.Then the variety of PI and ER expression of cholangiocytes by immunohistochemistry and variety of estradiol serum levels were determined in all groups.[Results]1.The collagen fiber morphology and structure of extrahepatic bile duct in human is similar to that in rat. Collagen fiber mainly distributes in middle and outer layer of bile duct. Collagen fiber bundles appeared wave-like and parallel, and there were connected branches between them which were in consistent with longitudinal axis direction of bile duct.2.ER is expressed in cholangiocytes of hepatohilar bile duct of SD rats. Expression of PCNA and ER of cholangiocytes in BDL group is apparently higher than that in control group. Expression of PCNA of cholangiocytes in BDL treated with tamoxifen group is apparently lower than that in BDL group. Estradiol serum levels of BDL group is higher than that of control group,while that of BDL treated with tamoxifen group is higer than BDL group.[Conclusions]1. The collagen fiber morphology and structure of extrahepatic bile duct in human is similar to that in rat. Anatomic structure is suitable to its function.2.Estrogen play an important role in BDL-induced proliferation of cholangiocytes,and antiestrogen tamoxifen can inhibit the effect of estrogen.
