Objective To investigate the effect of donor bone marrow cells transfusion on inducing pancreatic allograft tolerance in rat model. Methods SD male rats were used as bone marrow cells and pancreas donor , and BN female rats as diabetic recipients. BN rats with streptozotocin(STZ)-induced diabetes mellitus (DM)were conditioned by three methods:GrouP1:only Pancreas transplantation(PT) were performed ;GrouP2:Cyclophosphamide(150mg/ kg ) was injected into abdomen of diabetic recipients for one day after PT; GrouP3:2. 0×108 donor bone marrow cells were injected into portal vein on the day of PT; GrouP4:2. 0×108 donor bone marrow cells were injected into portal vein on the day of PT and Cyclophosphamide was injected into abdomen of diabetic recipients for one day after PT . recipient rats were detected for specific segment of the Y chromosome in the peripheral blood lymphocyte on day 14 after PT by polymerase chain reaetion(PCR)and donor origin cells in the spleen by flow cytometry (FCM).Then, Pancreas allograft survival was observed. Results SRY gene and donor origin cells were detected in Group 4,but the other three groups weren't. Pancreas allograft survival of group 4 was significantly prolonged (18±2.2 days) compared with group1. group 2 and group 3 did not improve allograft survival. Conclusion donor bone marrow cells injected into portal vein on the day of transplantation and Cyclophosphamide injected into abdomen of diabetic recipients for one day after transplantation have strong synergism in inducing long term graft survival in this rat model.It relates to chimera and immune tolerance induced by bone marrow cells.
