Objective: The purpose of this study was to investigate the expression of endothelin-1(ET-1) and nitric oxide synthetase-3(NOS-3) in atrial tissue of acute atrial fibrillation in canine,and dicuss the relationship between the Prethrombotic state and the injury of endothelial system in atrial fibrillation.Method: Forteen adult mongrel canines were randomly divided into two groups: the sham control group and the acute atrial fibrillation group.Acute atrial fibrillation model were induced by fast atrial pacing,fast atrial pacing and fast transfuse or fast atrial pacing and intravenous acetycholine.The research process were devided into three parts: first, the model of canine acute atrial fibrillation were develop, second, by HE stain the tissue structural of the atrial were studied by light microscope, then performed Immunohistochemistry to detect the expression of endothelin-1 and nitric oxide synthetase-3 in atrial tissue.Result: 1.Acute atrial fibrillation model can be induced succeedly in any of the experiment group; 2.Compared with the sham control group ,the endothelium has been injuried in the acute atrial fibrillation group, and we can also found endothelial cell loss in the acute atrial fibrillation group which we can not found in the sham group; 3.Immunohistochemistry study indicated that in the acute atrial fibrillation group and the sham group,the expression of ET-1 were obviously stronger in the former, but the expression of NOS-3 were significantly lower in the former.Conclusion: Acute atrial fibrillation can induced tissue structural change in the atrial endothelium displayed as ET-1 high express and NOS-3 lower express, which indicate that there has endothelium injuried in the early atrial fibrillation, and this maybe one of the most possible mechanism of Prethrombotic state in atrial fibrillation.
