Objective:To detect the chromosomal aberration and genomic alterations in lung adenocarcinomas and to evaluate the relationship between the chromosomal aberration and development of lung adenocarcinomas.Methods:M-FISH technology was used to detect chromosomal numerable and structural aberrations in three lung adenocarcinoma cell lines.CGH technology was used to analyze the global genomic aberration in the fresh cancer tissue cells of 50 patients with lung adenocarcinoma.Results:M-FISH reveals numerous complex chromosomal rearrangements.Translocations between chromosomes 5 and 11 was observed in three of the 3 cell lines,translocations between 1 and 6,17 and 19 were observed in two of the 3 cell lines with a further 11 translocations translocations being observed in one cell line sample. Chromosomes 5,6,11,12,17 were most frequently involved in interchromosomal translocations.CGH revealed regions on 1q,2p,3q,5p, 5q,7p,8q,11q,12q,14q,16p,17p,19p,20q,21q and 22q to be commonly over-represented and regions on 2q,3p,4p,5q,7q,8p,9p,13q,14q,17p to be under-represented.The most common gains were found in 16p 13(50%), the most common lossed were found in 8p22(38%).16p13 amplification was observed to be associated with relative poor differentiation and late stage.Conclusion:M-FISH and CGH could be used as a powerful tool for identification of genomic alteration for lung cancer,and the above-mentioned genes may represent potential candidate genes in the study of lung adenocarcinomas pathogenesis and diagnosis.Further investigation of the recurrent aberrations will be necessary to identify the specific breakpoints involved and any role they may have in the aetiology, diagnosis and prognosis of lung adenocarcinomas.
