Objective: To establish the rat model of metabolic syndrome and primary explore the possible mechanism of renin angiotensin system in injury of heart and kidney with metabolic syndrome by compare the difference of expression of ACE2 and ACE in heart and kidney of metabolic syndrome.Methods: Twenty adults male Wistar rats of specific pathogen free animal were groupped as follows: control group( A , n=10) and metabolic syndrome group( B, n= 10). Group B were treated with 30% fructose in drinking water to establish metabolic syndrome. Systolic blood pressure(SBP) was measured by tail-cuff method at the 0th and 6th weeks. At 6th weeks, Lee index, body fat, oral glucose tolerance(OGTT), triglyceride(TG), low-density lipoprotein cholesterol(LDLC) ,high-density lipoprotein cholesterol (HDLC ), total cholesterol (TC), insuline sensitivity index(ISI), free fatty acids (FFA), plasma, heart and kidney AngII were measured respectively. The level of expression of ACE2 and ACE were determined by RT-PCR and immunohistochemistry respectively. We observe the histomorphological changes of heart and kidney by light microscopic with HE staning.Results: (1)SBP, TG, triglyceride(TG), low-density lipoprotein cholesterol(LDLC) body fat and Lee index were found to be significantly increased in MS group compared with control group , But ISI, free fatty acids (FFA) and high-density lipoprotein cholesterol (HDLC ) were decreased in MS group as compared to control group (P<0.001) .Obtained success with made rat model of metabolic syndrome. (2)ACE2mRNA in heart and kidney were found to be significantly decreased (P< 0.001) and ACE mRNA in these tissue were increased in MS group compared with those in control group (P<0.001); There were no significant difference on level of ACE2 and ACE in both MS group and control group (P>0. 05).(3) ACE2 protein were stained along renal cortical tubules and myocardial cell. ACE2 protein expression in heart and kidney were lower by optical density method of immunohistochemisty in MS group than in control group(P<0.001); There were no significant difference on level of ACE2 protein between heart and kidney in MS group(P>0. 05).(4)The concentration of plasma and part tissue AngII were found to be significantly increased in MS group as compared to control group (P<0.001). (5) There were no significant difference on histomorphological of heart and kidney by light microscopic with HE staning.Conclusion: (1) Treated 30% fructose drinking water could build with characteristic for hypertension of model rats with MS. (2)In metabolic syndrome exist the change of renin angiotensin system. (3)The damage in tissue of MS were related to the decreased expression of ACE2.
