Objective To observe the effect of basic fibroblast growth factors(bFGF) on apoptosis and matrix metabolism of rat intervertebral disc cell cultured in vitro.Methods Rat intervertebral disc cells were cultured in vitro in monolayer.The well-grown intervertebral disc cells of second generation were induced by anti-Fas antibody to apoptosis,treated with bFGFs of different concentrations(10,100,1 000μg/L).By using flow cytometry(FCM),the apoptosis of the cells stained with Propidium Iodide(PI)was Assayed.The level of hydroxyproline and proteoglycan in culture medium were detected with chloramines T method and DMB chromometry method.Results①Compared with control group,the apoptotic ratio of the intervertebral disc cells caused by different concentrations(10,100,1 000μg/L)of bFGFs had a significant difference(87.62±11.06%,53.30±9.23%,16.75±4.21%; P<0.01).②The content of hydroxyproline and proteoglycan in bFGFs culture medium was significantly increased as the increase of concentration of bFGFs[hydroxyproline (6.71±0.33),(9.12±0.41),(11.58±0.12)μg/L,P<0.01;proteoglycan(23.21±2.87), (32.45±5.23),(37.18±3.22)μg/L,P<0.01].The concentration of bFGFs was positively correlated with hydroxyproline and proteoglycan.(ra=0.972,P<0.01; rb=0.907 P<0.01).Conclusion bFGF can inhibit the apoptosis and promot collagen and proteoglycan synthesis of rat intervertebral disc cell.
