Object:Investigating the effect of the TGF-?1?bFGF of Bu Gan Jian Yao Fang on the degeneration of lumbar intervertebral disc in rats.Methods:A total of 54 3-4 months old clean-grade healthy SD male rats were randomly selected from the lumbar disc herniated puncture method to prepare the lumbar disc degeneration model.After successful modeling,they were randomly divided into: Bu Gan Jian Yao Fang Low dose group,Bu Gan Jian Yao Fang middle dose group,Bu Gan Jian Yao Fang high dose group?model group,12 rats in each group,respectively,givenBu Gan Jian Yao Fang waist low dose(including crude drug 1.04g),Bu Gan Jian Yao Fang waist medium dose(including 2.08 g of crude drug),groupBu(including 4.16 g of crude drug)?normal saline,.The remaining 24 rats,12 in the normal control group and the sham-operated group,were not treated in the normal control group.The sham-operated group only exposed the intervertebral disc and did not undergo lumbar disc puncture.Four rats were sacrificed in each group at 2 weeks,4 weeks,and 6 weeks after administration.The tissue of the intervertebral disc degeneration segment L4/5?L5/6 was taken.After staining with HE,the morphology of the intervertebral disc tissue was observed by a 200 x optical microscope,and the content of TGF-?1 in the L4/5 intervertebral disc tissue was detected by immunohistochemistry(IHC).Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the expression of bFGF mRNA in L5/6 intervertebral disc tissue.Results:1.The rat lumbar intervertebral discs under the microscope were observed to show hyperplasia and degeneration morphological changes before drug intervention.After the drug intervention,the rat lumbar intervertebral disc tissue was repaired and improved.2.Rat lumbar intervertebral disc after modeling The expression of TGF-?1 and bFGF in tissues increased,and the expression of TGF-?1 and bFGF in rat lumbar intervertebral disc tissue decreased significantly after drug intervention,indicating that Bu Gan Jian Yao Fang can down-regulate lumbar intervertebral disc degeneration.The expression of bFGF mRNA and TGF-?1 can promote the repair of degenerative intervertebral disc.Among them,high and medium dose medicine decoction has no significant difference in this effect,but it is obviously stronger than low dose group.Conclusion:1.The lumbar intervertebral disc degeneration model can be success-fully made by acupuncture rat lumbar intervertebral disc.2.Lumbar disc degeneration is associated with bFGF and TGF-?1.3.Buganjian Yaofang has the effect of lowering the expression of bFGF and TGF-?1,which can promote the repair of degenerative intervertebral disc.The high and middle doses of Buganjian Yaofang group down-regulate the expression of bFGF and TGF-?1 in degenerative lumbar disc tissue.The effect of the lower dose group was significant and there was a significant difference.
