Pyritum has been combined with traditional Chinese medicine as fracture healing for a long time.Pyritum and some compounds with pyritum have many curative effects.It is for oral and external use.It is very important in traditional Chinese medicines.But the type of insoluble rough mineral has led to overdose of pyritum in drugs and insolution of active components.So it is restrict in medicine market.The treatment of pyritum with Acidthiobacillus ferriixidans(At.f),which was isolated and identified in our laboratory,was performed.Then we also did some initial research for the effects of pyritum leachate on the proliferation and differentiation of osteoblasts.And this paper studied some pharmacological effects of pyritum leachate on fracture healing and osteoporosis.From the changes of Fe,Cu,Mn,Zn,Se concentrations in pyritum leachate,the results demonstrated:the pyritum could be leached by At.f.The Fe concentration reached 1592μg/mL in 15-day,the extraction ratio was 68.33%.Cu,Mn,Zn,Se concentrations were increased than before bioleaching.MTT,PNPP were used to observe the effects of pyritum leachate on the proliferation and activity of ALP of osteoblasts cultured in vitro.The results demonstrated:the pyritum leachate had effects on inhibiting the proliferation in 1/40,1/80 concentration.The pyritum leachate had effects on stimulating the activity of ALP in 1/2560 concentration.This paper studied the effects of pyritum leachate on osteoporosis in rats.SD rats were randomly divided into groups,the serum Ca concentration,activity of ALP,BMD,BMC and histological sections were assayed after 6-week.The results demonstrated:the serum Ca concentration,BMD and BMC had no difference,but the activity of ALP was higher in the experimental group.The histological sections demonstrated:the trabecular bone structure was tighter in the experimental group,so we initially proved that the pyritum leachate had effects on osteoporosis in rats.The further research is studied.We also did some research for the effects of pyritum leachate on fracture healing in rats.SD rats were randomly divided into 3 groups,positive group,experimental group,negative group. The serum Ca concentration,activity of ALP,X-ray films and histological sections were assayed after 7,14,21-day.The results demonstrated:in the experimental group,the serum Ca concentration was obviously decreased than that in negative group after 14,21-day,and the activity of ALP was increased after 14-day,and the bone callus areas were obviously bigger than that in negative group after 14,21-day,and cartilage callus,trabecular bone and fracture healing were significantly earlier than that in negative group.
