Objectives:The aim of this study were to pinpoint whether single nucleotide polymorphisms(SNPs)in the genes coding for PECAM-1 were associated with ACI susceptibility in a Hunan Han population,and whether the SNP of PECAM-1 site L125V was associated with the plasma sPECAM-1 level.Method:142 subjects with ACI and 116 controls were enrolled in the study.Genomic DNAs were extracted from peripheral blood leucocytes by phenol-chloroform methods.The PECAM-1 L125V was genotyped by using restriction fragment length polymorphism method.The plasma sPECAM-1 level was measured by the ELISA method.Results:Stastically significant difference in PECAM-1 L125V genotypic distribution was observed between cases and controls(χ~2 =11.827,P=0.003).Moreover,the frequencies of the V allele was 59.5% in ACI group,and 47.9%in control group,prevalence of V allele in ACI subjects and controls were significant incomparable(χ~2 =6.998,P=0.008). Furthermore,the results of logistic regression analysis showed that the genotype of VV was associated with increase ACI risk(OR=2.355,95% CI:1.153-4.809,P=0.019.In both ACI group and control group,the plasma sPECAM-1 level of carriers of VV genotype at polymorphic site PECAM-1 L125V is higher than carriers of LL and LV genotype.The plasma sPECAM-1 level is associated with ACI.Conclusion:Our study showed PECAM-1 L125V genetic polymorphism is possibly associated with ACI.Carriers of VV genotype at polymorphic site PECAM-1 L125V showed increased risk for ACI. The PECAM-1 genetic polymorphism may associated with the plasma sPECAM-1 level.The plasma sPECAM-1 level is associated with ACI.
