Aim:1.To detect the expression of Smac/DIABLO and Caspase-3 in normal ovaries, benign ovarian tumors,borderline ovarian tumors and ovarian carcinomas,as well as study their association with clinicopathologic factors in ovarian carcinoma.2.To detect the expression of Smac/DIABLO in ovarian carcinoma cell lines of SKOV3 and SKOV3/DDP,and assess the relationship between Smac/DIABLO and chemoresistance of ovarian cells.Method:1.The representative paraffin sections were selected and 30 specimens of normal ovaries,30 specimens of benign ovarian tumors,30 specimens of borderline ovarian tumors and 60 specimens of ovarian carcinomas were tested by immunohistochemical technical with antibody against Smac/DIABLO and Caspase-3.2.To observe the expression of Smac/DIABLO in ovarian carcinoma cell lines of SKOV3 and SKOV3/DDP by RT-PCR.Results:1.Smac/DIABLO was found in normal ovaries,benign ovarian tumors,borderline ovarian tumors and ovarian carcinomas.The Smac/DIABLO in normal ovaries was significantly higher than that in borderline ovarian tumors and in ovarian carcinomas(x~2=8.52,P<0.0083;x~2=19.15,P<0.0083).The expressions of Smac/DIABLO in benign ovarian tumors was significantly higher than that in ovarian carcinomas(x~2=14.06,P<0.0083).2.In ovarian carcinomas,the expression of Smac/DIABLO was significantly decreased during progression from early to advanced tumour clinical stages and pathologic stages(F=7.23,P<0.05;F=4.48,P<0.05).The Smac/DIABLO level was significantly lower inⅢ-Ⅳstage than inⅠ-Ⅱstage ovarian tumors(t=2.35, P<0.05).The expression of was not significantly different in the different histological types of ovarian carcinomas,different ages and with ascitic fluid or not(P>0.05).3.Caspase-3 was found in normal ovaries,benign ovarian tumors and ovarian carcinomas.The Caspase-3 in normal ovaries was significantly higher than that ovarian carcinomas(x~2=12.86,P<0.0167).4.In ovarian carcinomas,the expression of Caspase-3 was significantly decreased during progression from early to advanced tumour clinical stages and pathologic stages(F=6.33,P<0.05;F=10.07,P<0.05).The Caspase-3 level was significantly lower inⅢ-Ⅳstage than inⅠ-Ⅱstage ovarian tumors(t=3.78,P<0.05).The expression of Caspase-3 was not significantly different in the different histological types of ovarian carcinomas,different ages and with ascitic fluid or not(P>0.05).5.In ovarian carcinomas,the expression of Smac/DIABLO was significantly correlated with Caspase-3(r=0.57,P<0.05).Smac/DIABLO and Caspase-3 coexisted in different ovarian tissues.Smac/DIABLO and Caspase-3 were observed almost in the same tissue and region.6.The patients with higher expression of Smac/DIABLO had favourable prognosis than those with negative expression(P<0.05).7.Smac/DIABLO mRNA was detected in ovarian cell lines SKOV3 and SKOV3/DDP,the relative content of Smac/DIABLO mRNA in SKOV3 was higher than that in SKOV3/DDP.Conclusion:1.The expressions of Smac/DIABLO in ovarian carcinomas are lower than normal ovarian tissue and benign ovarian tumors.The expressions of Caspase-3 in ovarian carcinomas are lower than normal ovarian tissue2.In ovarian carcinomas,the expression of Smac/DIABLO and Caspase-3 are significantly decreased when the clinical stages and the pathologic stage advanced. Thereby Smac/DIABLO and Caspase-3 play an important role in the occurrence and the development of ovarian cancer.3.The expression of Smac/DIABLO is significantly correlated with Caspase-3 which demonstrate that Smac/DIABLO maybe promote the Caspase-3 apoptosis way. 4.The low level of Smac/DIABLO was correlated with the poor prognosis of ovarian carcinomas.5.Smac/DIABLO was detected in ovarian cell lines and had relationship with the chemosensitivity to cisplatin in ovarian carcinomas.
