Expression And Clinicopathological Significances Of MCP-1, Macrophage, VEGF And MMP-9 In Gastric Carcinoma | | Posted on:2009-03-02 | Degree:Master | Type:Thesis | | Country:China | Candidate:X F Wang | Full Text:PDF | | GTID:2144360245484131 | Subject:Pathology and pathophysiology | | Abstract/Summary: | | | ObjectivesThrough detecting the expression of monocyte chemoattractant protein-1(MCP-1),vascular endothelial growth factor(VEGF)and matrix metalloproteinase-9(MMP-9)in gastric cancer and the counts of the infiltration of tumor-associated macrophages TAMs in gastric cancer stroma,to study their correlation with the tumor carcinogenesis,development,metastasis and angiogenesis.MethodsMCP-1,TAMs,VEGF,MMP-9,MVD were detected by immunohistochemistry streptavidin-peroxidase method(S-P method)in 20 gastritis and 50 differentiated gastric cancer.To analyze their dependability and the relationship with ciinicopathological parameters in gastric cancer.Results1.MCP-1 expressed both in gastritis and gastric cancer,but the expression level of MCP-1 in gastric cancer was higher than gastritis(P<0.01).there were no statistically significant of the MCP-1 expression in histology grouping and differentiation(P>0.05).And MCP-1 expression was concerned with the metastasis of gastric cancer(P<0.05).2.The counts of macrophage in gastric cancer were higher than their in control group (P<0.01).there were no statistically significant of the macrophage counts in histology grouping and differentiation(P>0.05).And the macrophage counts was concerned with the metastasis of gastric cancer(P<0.05).3.The MVD in gastric cancer were higher than them in control group and the difference was significant on statistic(P<0.01).And MVD with lymphoid node metastasis group were higher than them with no lymphoid node metastasis group(P<0.05).But differences of MVD in different differentiated degree of tubular adenoma and different typing of gastric cancer had no ststistical significance(P>0.05). 4.The expression level of VEGF in control group was lower than that in gastric cancer significantly(P<0.01).The expression of VEGF was correlated with the typing of gastric cancer,the expression level in intestine type gastric cancer was lower than suffusion gastric cancer(P<0.05),but had no relation with the differentiation.The expression level of VEGF with lymphoid node metastasis group were higher than them with no lymphoid node metastasis group(P<0.05).5.The expression level of MMP-9 in gastric cancer was higher than that control group,and the difference had the statistical significance(P<0.05).The expression of MMP-9 was concerned with the differentiation and typing of gastric cancer,the MMP-9 level in highly and moderately differentiated tubular adenocarcinoma was lower than poorly differentiated tubular adenocarcinoma(P<0.05)and the expression of MMP-9 in intestine type gastric cancer was lower than suffusion gastric cancer(P<0.05).Moreover,The expression level of VEGF with lymphoid node metastasis group was higher than that with no lymphoid node metastasis group(P<0.05).6.The expression of MCP-1 in gastric cancer had positive correlation with VEGF and MVD,but the dependability with MMP-9 had no statistically significant(P>0.05).The macrophage counts with the expression of MCP-1,VEGF and MMP-9 all had the positive correlation.Furthermore,VEGF expression had a positive trend,and the same with MMP-9.Conclusion1.MCP-1 could be produced by both tumor cells and macrophage,also could attract specifically more macrophage to tumor site.MCP-1 and macrophage promoted synergistically the progression of gastric cancer.2.MCP-1 might be an important angiogenesis factor in cancer.On the one hand,it could up-regulate the expression of VEGF to promote angiogenesis,on the other hand,MCP-1 could attract macrophage into the tumor site and macrophage can produce many angiogenesis factors to promote the tumor angiogenesis.3.The more MCP-1 expressed,the easier lymphoid node metastasis was happened.
