Objectives:To establish animal models of human prostate cancer,and then treat with TGF-βinsensitive and tumor-reactive CTL;Collect xenografts and use to some experiment,such as extraction of TAA and determination of TGF-βlevel;Culture the cancer cell from xenografts and compare with other primary culture method,and develop a more potential one.Methods:Using immunodeficient mice(4 week-old,male)to establish xenografts animal models from patients with prostate cancer.When the diameter of xenografts reach 1.5cm,sacrifice the mouse and make another models.At last,we have done 3 implantations,and obtain mice with xenografts.Most of them are divided into 2 group and treated by CTL through the caudal vein,collect and analyze the date with statistical software;Collect xenografts and use to extract TAA and determinate TGF-βlevel and to proliferate the CTL;The rest of xenografts will be used to culture prostate cancer.Results:1.Use 12 animal models to make the study of treatment of TGF-βinsensitive and tumor-reactive CTL to prostate.cancer.The difference between treatment and control group had statistical significance.2.Xenografts from mice can provide enough prostate cancer tissue.TAA extracted from xenograft can proliferate the CTL obviously,it has statistical significance.The TGF-βlevel of serum is 4.873ng/ml.The TGF-βlevel of xenografts,12.116ng/ml is higher than the prostate tissue from patient,8.368ng/ml.3.Although we did not establish the prostate cancer cells line,Prostate cancer cells from xenografts have a more potential ability to proliferate than tissue block culture method.Conclusions:1.The study of treatment of TGF-βinsensitive and tumor-reactive CTL to prostate cancer has great clinical significance.And immunodeficient mice model is simple, economic and effective way to research the immunotherapy of prostate cancer.2.Xenografts can make up the insufficiency of prostate cancer specimens,it can provide enough cancer tissue in other research,such the extraction of TAA and determination of TGF-β.The higher level of TGF-βin xenografts demonstrate that it is significant to study the immunotherapy of prostate cancer though the way of TGF-β.3.Prostate cancer cells from xenografts have a more potential ability to proliferate than tissue block culture method.It is a good way to establish prostate cancer cell line.
