ObjectiveTo investigate the expression of IL-1β,TNF-αand TNFRⅡin invasive micropapillary carcinoma of the breast(IMPC),and analyze their relationship with the pathologic features and their role in angiogenesis in IMPC,then compare the expression of IL-1β,TNF-αand TNFRⅡwith that in invasive ductal carcinoma (IDC).MethodsPrimary pathologic features(ER,PR,Ki67,histological grade,lymph node metastasis and extranodal extension)of 100 cases of IMPC were examined by microscopic analysis.Immunohistochemical analysis for IL-1β,TNF-α,TNFRⅡand CD34 was performed,and microvessel density was measured on these cases.97 cases of IDC were chosen as control group.Results1.Tumor size,the number of positive lymph nodes and extranodal extension in IMPC were all much higher than that in IDC(P<0.05);2.Neither tumor size or the amount of IMPC in the tumor was associated with its lymph node metastasis(P>0.05),and the metastasis foci in lymph node were all IMPC or mainly IMPC(P<0.05);3.The expression of IL-1βhad no significant difference between IMPC and IDC(P=0.924),and it was negatively correlated with ER(P=0.035),and positively correlated with Ki67(P=0.023),histological grade(P=0.038) and lymph node metastasis(P=0.008);4.The expression of TNF-αin IMPC was higher than that in IDC(P=0.045). In IMPC TNF-αexpression showed significantly positive correlation with proliferation of tumor cells(P=0.007),histological grade(P=0.003),lymph node metastasis(P=0.016);5.The expression of TNFRⅡwas much higher in IMPC than that in IDC (P=0.022),and it was correlated with TNF-αexpression(r=0.399,P=0.000) and the proliferation of tumor cells in IMPC(P=0.029);6.The MVD in IMPC was significantly higher than that in IDC (66.38vs59.96,P=0.003).The mean MVD in node-positive IMPC was higher than that in node-negative IMPC(68.82vs54.44,P=0.001).It was not IL-1βbut TNF-αthat was more important in angiogenesis in IMPC.Conclusion1.IMPC was more lymphotropic than IDC;2.Neither tumor size nor the amount of IMPC in the tumor was associated with its lymph node metastasis;3.In IMPC,IL-1βexpression was also correlated with malignant phenotype, although it had no relationship with angiogenesis.This suggested that overexpression of IL-1βmay demonstrate highly invasive behavior;4.The expression of TNF-αand its receptor TNFRⅡwas related with the poor prognosis,and they played a role in tumor cell proliferation and angiogenesis in IMPC to enhance the aggressive behavior.
