Reflectance and fluorescence spectroscopies were applied in diagnosis and treatment of Photodynamic Therapy (PDT) for Port Wine Stain (PWS). With the object of PWS patients, the assessment for severity of Port Wine Stain (PWS), the detection of Epidermal Melanin Content (EMC) of PWS skin and oxygen saturation were studied.Firstly, a short introduction to study history of treatment for PWS and the application situation of reflectance and fluorescence spectroscopies in skin diseases were outline. The physiology of PWS, testing system, basic theory for reflectance and fluorescence spectroscopies were also given.Secondly, based on the analysis of PWS pathology, an optical model of PWS was developed, the reflectance and fluorescence spectra for different severity of PWS were obtained by Monte Carlo simulation. At the same time the reflectance and fluorescence spectra of patients with different severity of PWS were measured. Result of measured spectra agreed with that of simulated spectra. Study showed that reflectance and fluorescence spectroscopies could be applied to detect the severity of PWS or therapeutic efficacy of PDT for PWS.Thirdly, according to the diffuse reflectance model, accompanied with the HE dying, optical coherence tomography (OCT), we discussed a technique which can noninvasively measure EMC of human skin and the feasibility of using this technique to test the Epidermal Melanin Content(EMC) before or after the treatment of the PWS. Meanwhile we put forward another noninvasive measurement method about blood oxygen saturation in PDT for PWS in this research. All above research results could supply references of clinical therapeutic regimen and therapeutic efficacy of PWS.Finally, summarize the main work of the research and bring forward the object of the further study of PWS.
