Objective: To elucidate the functional status of DCs in the tissue of Renal cell carcinoma (RCC) and in the peripheral b1ood and to investigate its relationship with RCC escape from immunosurveillance.Methods:In this experiment ,40 specimens from RCC cases primarily treated with surgery were selected as test group. In addition ,20 specimens of normal Kidney tissue from kidney stone sufferers which part of kidney was cut off were used as control. Distribution of DC expressing CD1a+ , CD83 + and HLA-DR+ in tumor tissue and normal tissue was observed by immunohistochemistry. The density of DC and the rate of DC expressing HLA-DR in Renal cell carcinoma group and control were statistically compared.At the same time DC was Isolated and purified from peripheral blood of 20 renal cell carcinoma patients which were in hospital from 2006 to 2007 year from above mentioned 40 cancers and 20 health volunteers.DC was authenticated through cell droping slice and immunohistochemistry. The DC variety appearance was observed through light mirror and upside down microscope.DC was detected the expression level of CD1a and CD83 costimulatory molecular through flow type cell instrument.Result: The CD1a + DC and CD83+ DC was found in cancers. The density of CD1a+DC(3.2/HPF)and CD83+ DC(4.4/HPF)in tumor tissue was significantly lower than that in normal tissue(49.2/HPF)and(44.3/HPF)(P<0.05) . HLA-DR antigen expressed on the surface of DC in tumoral tissue of 34 cases carcinoma specimens and in normal tissue of 16 cases. The rate of HLA-DR positive expression had no statistic significance between the two groups. The expression level of CD1a and CD83 (57.4±1.6) and (72.1±1.1 ) on DC from RCC peripheral b1ood was lower than that(64.9±1.4)and (80.6±1.2)(P < 0.05)from contrasts.Conclusion:DC has the tendency to gathering in tumor. There were CD1a + DC and CD83+ DC in RCC tissue .But the expression level of CD1a + DC and CD83+ DC was lower than that from contrasts . The rate of HLA-DR positive expression had no statistic significance between tumor and normal control .The expression level of CD1a and CD83 on DC from RCC peripheral b1ood was lower than that from contrasts. The results indicate that the functional deficit of immune of the DC from RCC could be an important reason for tumor to escape from the immunology surveillance of tumor- bearing host.
