Objective To construct a recombinant expression vector bearing HBV C gene and encoding gene of hydrophobic structure domain ,and to explore the expression of fusion gene in membrane of Human hepatocellular liver carcinoma cell line(HepG2)cells.Methods Encoding gene of signal sequence of transferrin receptor(TfR)was recombined into 5' terminal of HBV C gene by overlaping primer design and polymerase chain reaction(PCR) amplification, then which was inserted into expression vector PCDB. The recombinant expression vector PCDB -Tfr–HBcAg has been transfect into HepG2 cells. The expression of fusion protein in cellular membrane of HepG2 cells was investigated by indirect immunofluorescence analysis.Results HBcAg with hydrophobic structure domain was detected in cellular membrane of HepG2 cells by immuno-fluorescence microscope.Conclusion The successful membrane expression of Hepatitis B core antigen might provide an experimental evident for HBcAg bearing target antigen of immuno-pathogenesis in chronic hepatitis B.
