Objective: To observe effects of Troglitazone on mRNA and protein expression of ICAM-1 in TNF-αcultured human mesangial cells, and to explore anti-inflammatory effect and potential mechanism of Troglitazone in human mesangial cells, which may provide pharmacologytarget and theoretical basis for future prevention and treatment of glomerulonephritis.Method: HMCL were divided into control group, TNF-αgroup, TNF-α+ Troglitazone group, and Troglitazone group.Proliferation measurement of each group with MTT,mRNA measurement of ICAM-1 in each group with RT-PCR,Protein measurement of ICAM-1 in each group with ELISA,Protein measurement of NF-κB in each group with ICC.Result: (1)Results of MTT : The proliferation of human mesangial cells in TNF-αgroup was significantly higher than in control group(P<0.05).The maximal proliferation level of mesangial cells stimulated by TNF-αwas present at the 24th hour which was significantly higher than the 6th hour group(P<0.05). Compared with TNF-αgroup, the proliferation of mesangial cells was significantly lower after adding different concentrations of Troglitazone(P<0.05).(2) Results of RT-PCR: The mRNA level of ICAM-1 in different concentration of TNF-αgroups was significantly higher than in control group(P<0.05). Compared with TNF-αgroup, the mRNA level of ICAM-1 was significantly lower after adding different concentration of Troglitazone(P<0.05).(3) Results of ELISA: The protein level of ICAM-1 in different concentration of TNF-αgroups was significantly higher than in control group(P<0.05). Compared with TNF-αgroup, the protein level of ICAM-1 was significantly lower after adding different concentration of Troglitazone(P<0.05).(4) Results of ICC: The protein positive rate of NF-κB in different concentration of TNF-αgroups was significantly higher than in control group(P<0.05). Compared with TNF-αgroup, the protein positive rate of NF-κB was significantly lower after adding different concentration of Troglitazone(P<0.05).Conclusion: (1) TNF-αcould stimulate mesangial cells proliferation, up-regulate mRNA and protein expression of ICAM-1 and protein expression of NF-κB, which suggested that TNF-αcan promote human mesangial cells inflammatory reaction.(2) Troglitazone could down-regulate the proliferation level of human mesangial cells stimulated by TNF-αand inhibit mRNA and protein expression of ICAM-1, which suggested that Troglitazone can inhibit human mesangial cells inflammatory reaction.(3) Troglitazone could down-regulate the protein expression of NF-κB in human mesangial cells stimulated by TNF-α, which suggested that Troglitazone may decrease the expression of ICAM-1 through inhibiting NF-κB activity .
