Objective: To investigate whether or not celecoxib can improve cisplatin-induced apoptosis in vitro.Methods:Osteosarcoma cell(MG-63)were cultured with celecoxib,cisplatin or celecoxib with cisplatin.We called them control group, celecoxib group, cisplatin group and mixing group respectively. The cells were examined by flow cytometry(FCM) and stained with acridine orange/ethidium bromide (AO/EB)to determine the morphology of the cells and the level of apoptosis in each group.The MTT assay was used to estimate the condition of cell growth.Enhancement effect of celecoxib in cisplatin-induced apoptosis of osteosarcoma was observed from three aspects.Results:Celecoxib alone can inhibit the proliferation of osteosarcoma MG-63 cells in a dose-dependant manner. Apoptosis were observed in three drug groups.The growth inhibition rate of MG-63 cells treated with 20, 40, 80μmol/l celecoxib were 8.07%, 40.30%, 56.50% at respectively (P<0.05,P<0.01). Celecoxib at concentration of 40 and 80μmol/l cause significant morphological changes:cancer cells were sparsely distributed and many slim processes in the edge of cells. Celecoxib can enhance the killing effects of cisplatin at low concentration(1.25,2.50μg/m1) and two drugs had reciprocal effects .The growth inhibition rate of MG-63 cells treated with 1.25, 2.50μg/ml cisplatin were 60.5%, 69.6% respectively (P<0.05,P<0.01). The growth inhibition rate of each mixing group:71.3% and 74.2% at cisplatin 1.25μg/ml + celecoxib 40μmol/l, and cisplatin 1.25μg/ml + celecoxib 80μmol/l,77.1% and 85.6% at cisplatin 2.50μg/ml + celecoxib 40μmol/l and cisplatin 2.50μg/ml + celecoxib 80μmol/l. Percentage of apoptotic cells in mixing group was higher than that's in other groups(P<0.05).Conclusion: Celecoxib and cisplatin can induce apoptosis of osteosarcoma cell line (MG-63)respectively, while the cell line treated with celecoxib and cisplatin simultaneously,the apoptosis rate was increased obviously.
