| Objective: To study the method of constitution DC vaccine modified by Hsp70-H22 tumor peptide complex,to observe the cytokine IL-10,IL-12 secreted by DC, the surface marker CD11c,CD40,CD80 expressed on DC and the antitumor effect in vitro .Methods:The tumor antigen peptides were acquired from H22 liver cancer cells and bound HSP70 in vitro by using biochemical technique; the mouse marrow cells were cultured with induction of rmGM -CSF and rmIL-4 by using cell culture technique,and obtain mass DC.Activate DC by Hsp70-H22 tumor peptide complex ( 10ug, 20ug ) separately. The concentration of IL-12 and IL-10 were measured in culture supernatants by ELISA. Analyse the surface marker CD11c,CD40,CD80 by fluorescence-activated cell sorter(FACS).Results: DCs could become mature with the effect of Hsp70-H22 pepfide complexes, and express CD40,CD80.the express rate of Hsp70-H22/DC group were distinctly increased than DC group(P<0.01),there were no difference between the groups(10ug,20ug) of Hsp70-H22/DC (P>0.05)。The concentrations IL-12 secreted by Hsp70-H22 tumor pepfide complexes modified DC were higher compared with DC,and IL-10 secreted by Hsp70-H22 tumor peptide complexes modified DC were lower compared with DC(P<0.05).There were significant statistical significance between (10ug ,20ug) groups(P<0.01).Conclusion: DCs origined from marrow cells can be effectively modified by Hsp70-H22 tumor peptide complexes , DC vaccine modified by Hsp70-H22 tumor peptide complex can improve the immune state , lead to up-regulation of CD40,CD80,increase cytokine of IL-12 and decrease IL-10, and improve the anti-tumor effect. |
