Part One Changes of super microstructure of hippocampus and expression of caspase-3 in rats induced by amyloid YVAD-CHOObjective To investigate the changes of super microstructure of hippocampus and expression of caspase-3 in rats reduced by amyloid YVAD-CHO.Methods Thirty-two female SD rats were randomly divided into control and model groups. YVAD-CHO was injected into bilateral hippocampus in model group.At the end of the experiment expression of caspase-3 protein were studied by immunohistochemistry method.Result1.The level of epileptic seizure reached Racineâ…£-â…¤2.By amyloid YVAD-CHO can lessen the frequency and level of seizure .3.By amyloid YVAD-CHO can lessen the cell damage.4.By amyloid YVAD-CHO can prevent the expression of caspase-3. Conclusion By immunohistochemistry, fewer hippocampal neurons in CA1 region were observed in model group, of which some crippled cell membranes, shrinked cell bodies, and enhanced electric densities of chromatin were found. By HE staining, much fewer hippocampal neurons in CA1-CA3 regions with condense chromation in nucleus were observed. The expression of caspase- 3 protein in hippocampus highly increased in model group. Conclusion: YVAD-CHO can reduce apoptosis of rat's hippocampal neurons in vivo. Part Two Empirical study the PET appearance of YVAD-CHO interfere in epilepsy rabbitsObjective To show the PET appearance of YVAD-CHO interfere in epilepsy rabbits.Methods To set up the model of epilepsy rabbits and YVAD-CHO intervention model. To gain the interparoxysmal imageology appearance of PET/CT .Result To set up the model of epilepsy rabbits and YVAD-CHO intervention model.Conclusion The interparoxysmal imageology appearance of PET/CT can improve that the metabolism of 18F-FDG of the rabbit which has injected YVAD-CHO is higher than the one did not.
