Traditional Chinese medicinal materials HPLC fingerprint technology is the macro-analysis of the complex chemical characteristics information by use of high-performance liquid chromatography, which reflects the differences in genetic species and individual according to the integrity and fuzzy principles by qualitative and quantitative method. It is an important content in traditional Chinese medicinal materials quality control promoting the modernization and universal application of Chinese medicine development and having positive significance.Firstly we take Platycladus orientalis(L.)Franco from Emei Mountains as an example to explore separation conditions, including the mobile phase, detection wavelength, column, flow and temperature, the sample volume and the gradient elution procedures, analyzing the quercetin content influence of the different processing methods to emphasize the importance of controlling sources in the establishment of the fingerprint, secondly we take Agrimonia pilosa Ledeb., Prunella vulgaris L. and Euphorbia humifusa Willd. as examples to analyze the conditions for optimization in fingerprint establishment, especially the steep and width choice in gradient elution procedures, illustrating the general process of separation conditions optimization under the guideline of the different parts of traditional Chinese medicinal materials and the different content chemical composition.This article initially explore the traditional Chinese medicinal materials HPLC fingerprint establishing methods, bringing forward the framework of construction for traditional Chinese medicinal materials HPLC fingerprint data, at the same time as a part of the research projects Biological samples integration and sharing in nature reserve, providing an initial basis and reference data for the latter part of the Emei Mountains traditional Chinese medicinal materials standard chromatographic fingerprint establishment and evaluation.
