Tanggut rhubarb is the dried root and rhizome of Rheum tanguticum Maxim.ex Balf.,which is one of the authentic rhubarbs included in the Chinese Pharmacopoeia.The genuine producing area of Tanggute Rhubarb is Qinghai Province and Qilian Mountain in Gansu Province.Angelica is the dry root of the perennial herbaceous plant Angelica sinensis(Oliv.)Diels.The genuine producing area of Angelica sinensis is Minxian and surrounding counties in Dingxi City,Gansu Province.Angelica sinensis produced in this area is also called "Mingui".The main purpose of this article is to consolidate the advantages of these two kinds of the genuine medicinal materials in Gansu Province,comprehensively evaluate the quality of Tanggut rhubarb and Angelica sinensis,and understand the processing methods of the origin tablets of these two kinds of medicinal materials.The thesis is divided into two parts.The first part is to understand and compare the processing methods of Tanggut rhubarb production and comprehensive evaluation of the quality of Tanggut rhubarb;the second part is to understand and compare the processing methods of Angelica sinensis production,evaluate the quality of Angelica sinensis and distinguish different Angelica sinensis from different origins.The first part focuses on the processing and quality evaluation of Tanggut rhubarb producing tablets.The main work of this part is as follows: 1.Summarize and compare the processing methods of rhubarb and Tanggut rhubarb producing tablets,and recommend low-temperature drying or natural drying as suitable methods of drying.Then briefly describes the processing methods flow of Tanggut rhubarb with different specifications and different grades.2.High performance liquid chromatography(HPLC)was used to determine the content of anthraquinones,phenolic acids and dianthrones in many batches of Tanggut rhubarb samples,and the regular patterns were summarized and compared.3.Combined with the content of the medicinal ingredients determined in item 2,the principal component analysis method was used to evaluate the quality of the longitudinal distribution of each part of Tanggut rhubarb from the main root to the branch root and The quality of each part of the lateral distribution of the five-year main root and branch roots was further compared.Comparing with the quality of main roots and branch roots in root systems of different years and it was concluded that the quality of rhubarb will increase with the growth of years.The main component score of four-year-old Tanggut rhubarb reached the peak,and its quality is the best as well.4.The discriminant analysis method was used to analyze Tanggut rhubarb of different years,and an effective discriminant model was established to judge the growth years of Tanggut rhubarb.The second part focuses on the processing and quality evaluation of Angelica's production.This part mainly does the following work: 1.Summarize the various methods of processing Angelica sinensis tablets,compare the advantages and disadvantages of each method,and recommend the natural drying method as a suitable processing method.2.The content of basic inspection items such as water,ash,acid-insoluble ash,Leachate,volatile oil and the content of ferulic acid and ligustilide in Angelica sinensis by different drying methods were determined and compared.3.Established a fingerprint detection method for the methanol extracted components,water extracted components and volatile oil components of Angelica sinensis,and established the fingerprint spectra of each component.And based on the main peak area of the fingerprint spectrum,cluster analysis was carried out to achieve the purpose of comprehensive evaluation of the quality of angelica.4.The infrared spectroscopy(IR)was used to collect the mid-infrared spectra of angelica from different origins,and the PCA-MD model was established using AQ Analyst 8.3.The accuracy rate was 99.3%,which can accurately determine whether the Angelica sinensis sample is Mingui.The PLS-DA model was established using SIMCA 14.1.The R2 Y value was 0.946 and the Q2 value was 0.812.It was verified that the model was stable,effective,and without overfitting,which could effectively determine the origin of the Angelica sinensis samples.
