Purpose The pathological proliferation of fibroblast was interfered by connective tissue growth factor antisense oligonucleotide(CTGF ASODN)through the mediating with liposome after glaucoma filtering surgery (GFS) in rabbit model. To get further understanding of the role of CTGF, observe the the influence of CTGF ASODN and discuss the effect of CTGF ASODN mediating with liposome in the scar formation that occurs after glaucoma filtering surgery (GFS) in rabbit model. And offer a new method of anti-scarring in filtering bleb area after GFS.Methods The tissue from conjunctiva and partial-thickness sclera of filtering bleb area was harvest and carried out primary culture in the fifth day after GFS in rabbit. CTGF ASODN conjugated with isothiocyananate fluorescence was encapsulated by liposome and transfected into fibroblast. The cytoplasmic distribution of CTGF ASODN was observed with fluorescencemicroscopy. And the expression of CTGF in mRNA and protein level was measured by RT-PCR and Western-Blot tests seperately. To observe the effect of CTGF ASODN with relative gray scale as the index.Results (1) A few fluorescence spots were found in the cytoplasm of research group at 6h. And large number of fluorescence spots were observed in the cytoplasm of research group at 12h after transfection separately. While there is little fluorescence spot in the control groups. (2) The mRNA index(RI) of CTGF was significantly depressed in the research group compared with that in the control group in mRNA level, RI was 0.457±0.035(P<0.01 vs D group).And the expression of CTGF was reduced at the same time, RI was 0.177±0.022(P<0.01 vs D group).Conclusions (1) Liposome can transfect CTGF ASODN effectively. (2) Culture in vitro suggested that CTGF plays an important role in scarring and wound contracture after GFS. And CTGF ASODN can inhibit the expression of CTGF in mRNA and protein level comparatively.
