Objective The reserch weather the expression and function of PI3K/Akt could be affected by siRNA(small interferingRNA) with multi-drug resistant leukemic cells HL-60/VCR.Methods HL-60/VCR cells were used,with a 19 bases sequence of siRNA aimed Akt1 gene and negtive sequence which had nothing to do with Akt1 gene.The first step was to transfect siRNA sequences into HL-60/VCR cells with liposome,then flow cytometry wss used to detect the different expression of the 473th p-Ser of Akt1 after siRNA towards Akt1,and the data of the expression of P170,CD123+ and CD34+ CD38- was also got.Last, the change of inhibition to VCR of the cells transfected after the treatment mentioned above by chemosensitivity assay in vitro(MTT).Results The four main indicators that of p-Akt1,p170,CD123+,CD34+CD38- in experimental group were declined significantly compared with control group and negtive group(p<0.05).Another result from chemosensitivity assay in vitro(MTT) shew that HL-60/VCR become more sensitive to VCR(p<0.05).Conclusion The siRNA targeting PI3K/Akt might inhibit p-Akt specifically in HL-60/VCR and make CD34+ reduce,which dewn the expression of CD34+,inhibited LSC(Leukemia Stem Cells) to a certain degree and made P170 declined.This way would make leukemic cells become more sensitive to chemotherapy and could have new ideals contribute to treat leukemic multidrug-resistance in clinic.
