Objective:To investigate the role of urinary protein from lupus nephritis patients on expression of NF-κB,TNFα,IL-6 in human proximal tubular epithelial cells(HK-2).Methods:Urinary protein was purified from the urine of patients with lupus nephritis and incubated with HK-2 cells. The HK-2 cells were divided into two groups: one group incubated with the urinary protein from lupus nephritis patients with different times(1h,3h,6h,12h,24h,48h), another with cultured with culture medium without serum 24 hours. The expression NF-κB,TNFα,IL-6 mRNA of HK-2 cells was assessed by reverse transcription- polymerase chain reaction(RT-PCR).Results:The urinary protein up-regulated the expression of NF-κB,TNFα,IL-6 mRNA in HK-2 cells. The expression of NF-κB,TNFα,IL-6 mRNA was associated with the stimulation time. The levels of NF-κB,TNFα,IL-6 mRNA was increased in time- dependent manner. The levels of NF-κB,TNFα,IL-6 mRNA was increased in the supernatant of culture at 48 hours. However, the expression NF-κB,TNFα,IL-6 mRNA of HK-2 cells cultured with culture medium without serum 24 hours was faintness and not increased. The level of NF-κB mRNA was closely positive correlated with TNFαmRNA and IL-6 mRNA.Conclusions:Urinary protein can up-regulated the expression of TNFα,IL-6 mRNA in HK-2 cells by NF-κB signal pathway. The levels of NF-κB,TNFα,IL-6 mRNA was increased in time-dependent manner within 48h.Those may play important roles on the proteinuria induced tubulointerstitial damage during progression of chronic glomerular disease.
