Objective(1) To observe the associatively between pathology and expression of CK19mRNA and CEA mRNA from peritoneal cavity washes of gastric cancerpatients.(2)To observe the clinical value of CK19 mRNA and CEA mRNA detected bynested RT-PCR for diagnosing celiac micro metastasis of gastriccancer cells.Methods(1)Peritoneal irrigating solution or abdominal dropsy was collectedfrom 67 patients with advanced gastric cancer and 8 patients withbenign lesion on stomach, and meanwhile, their peripheral blood wascollected.(2)Peritoneal irrigating solution or abdominal dropsy was stained byHE for cytological examination.(3)Peritoneal irrigating solution or abdominal dropsy was taken toobserve CK19 mRNA and CEA mRNA by nested RT-PCR detection.(4) Peripheral blood was taken to observe the expression of CEA proteinby chemoluminescence immunization. Gastric cancer cell line SGC7901 was served as positive control.Results(1)By nested RT-PCR detection, CK19 mRNA was positive expressed on 39of 67 patients(58.2%), CEA mRNA was positive expressed on 36 of 67patients (53. 7%), and CK19 mRNA or CEA mRNA was positive expressed on45 of 67 patients(67. 2%).(2)By cytological examination, Cancer cells were positive expressed on13 patients of 67 patients (19.4%), and in which both CK19 mRNA andCEA mRNA was positive.(3)There was a close correlation between the expression of CK19 mRNAand CEA mRNA and pathology and TNM staging for gastric cancer patients(P<0.05).(4)By chemoluminescence immunization, CEA protein was positiveexpressed on 23 of 67 patients (34.3%).(5)There was a close correlation between the expression of CEA protein and pathology and TNM staging for gastric cancer patients ( p < 0. 05) , but is was not a sensitive method for diagnosing celiac micro metastasis of gastric cancer cells.ConclusionsIt is sensitive to detect CK19 mRNA and CEA mRNA of peritoneal irrigating solution from gastric cancer patients by nested RT-PCR, and it is promising as a valuable method for predicting celiac implantation metastasis in gastric cancer.
