ObjectiveTo investigate sera glutathione antioxidation ability and the relationship with expression ofα-interferon receptor 2(IFNAR2) in peripheral blood mononuclear cells(PBMC) and hepatocyte apoptosis of patients with chronic hepatitis B(CHB). This experiment can guide clinical works for the future and can extend in the clinicMethodsThe controls include 5 females and 5 males whose mean age were 29.7±5.8 years (range,22-38 years). These selected normal persons have not any disease. The subjects were 66 patients with CHB, including 37 males and 29 females, age averaging (32.5 + 10.5) years (range, 16-51years). The patients all had been admitted to Qilu hospital of Shandong University and accorded with diagnosis criterion publicized at 2000. These selected patients have not other diseases. Sera glutathione(GSH), glutathione S-transferases(GST), glutathione peroxidase(GPX) and glutathione reductase(GR) are testd by chemic colorimetry. IFNAR2 mRNAs from PBMC were measured by reverse transcriptionpolymerase chain reaction(RT-PCR). Hepatocyte apoptosis of hepatic tissue was determined by tunel method. t - test was used for the intergroup comparison and the correlation test was undertaken with the linear correlation analysis.Results1. The concentration of sera GSH, GPX and GR of the patients is significantly lower than the controls. And the concentration of GST of the patients in serum is significantly higher than the controls. In CHB patients, the level of ALT has a significantly negative correlation with sera GSH and GPX, and has a significantly positive correlation with GST.2.The concentration of sera GSH and related enzymes has no statistically difference between HBVDNA positive and negative patients, and in CHB patients all enzymes have no significantly correlation with HBVDNA in blood.3. In CHB patients, expression of PBMCs IFNAR2 mRNA has no statistically significant difference with the controls. In HBVDNA negative patients, expression of PBMCs IFNAR2 mRNA is higher than positive patients. And the expression of PBMCs IFNAR2 mRNA has no significantly correlation with GSH and related enzymes in blood of CHB patients.4.Apoptosis index (AI)has no significantly correlation with GSH and related enzymes in blood of CHB patients.Conclusion1. Anti-oxidant defense decreased in subjects with chronic HBV infection and CHB patients suffered oxidative stress, and HBVDNA load can not influence oxidative stress.2. In CHB patients the expression of PBMCs IFNAR2 and apoptosis index have no significantly correlation with oxidative stress.3. As interactions existed among GSH and related enzymes, treatment of CHB patients with antioxidants should not contain GSH only.
