Objective:Pulpitis is one of the most common oral diseases, most of pulpitis caused by caries、 bacterial infection 、ischemia and hypoxia of pulp. GPx1 and GR are important enzyme to organism antioxidation, the physiological function and pathogenic mechanism in pulp tissue and their relationship with pulpitis are not clear at present, the experiment on their expression in dental pulp tissue,and their differences of expression in dental pulpitis were analyzed. Trying to analysis the antioxidant mechanism and antioxidant capacity of dental pulp cells from the level of cell and molecular level, so as to provide basis for clinical application of antioxidant drugs in clinical application. Method:According to the selection criteria to select 25 cases of normal, acute pulpitis and chronic pulpitis samples, all the specimens need the progress of fixation, embedding, sectioning, HE staining, according to pathology, divided into three groups, each group select 20 cases from the 25 cases befor, after the specimens by immunohistochemical staining, using optical microscope to observe the expression of GPx1 and GR in three groups of specimens. The positive rate of cells was calculated and the difference was analyzed. Result:The cell nucleus and / or cytoplasmic of the dentin cells, macrophages, vascular endothelial cells have brown granules in normal dental pulp, and a large number of neutrophils、 lymphocytes、 monocytes have been stained of dark brown in the acute dental pulp group, and in chronic pulpitis group,lymphocytes、monocytes、 macrophages were dark brown staining. The positive rates of GPx1 in each group were 54.82±7.90、69.93±6.50、63.05±4.67, the positive rate of GR respectively was 56.13±5.96、70.30±9.25、64.19±6.37.There were significant differences among the three groups(P<0.05). Conclusion:In normal dental pulp tissue, GPxl and GR as antioxidant enzymes, is widely expressed in tissues, especially odontoblasts, but it doesn’t express in fiber cells. This result suggests that the dentin cells involved in the body’s normal response to oxidative stress. The expression of GPxl and GR in odontoblasts and inflammatory cells is more stronger than normal groop, such as macrophages、 neutrophils、plasma cells、lymphocytes and so on. In the acute pulpitis, GPxl and GR expression were higher than that of chronic pulpitis, it’s maybe relate to the inflammatory cell damage in antioxidant system. The expression of GPxl and GR in all three group have significant difference.
