ObjectiveThere were varieties of microbes living in the gastrointestinal tract of healthy people,and the microbes are so called intestinal flora.Under the ordinary status,the intestinal flora was in a balance with the human body and its external environments,and played an important role in physical fitness.But there has had a lot of reports referred that overdose of antibiotics would cause the alteration of intestinal flora.According to the Chinese traditional medical theory,large dosages and/or long term usage of heat-cleaning and detoxifying traditional Chinese medicine will injure the spleen-qi and waste the healthy energy,at the same time,here were also some reports indicating that the bitter cold medicines,like the Coptidis Decoction for Detoxification,had the effect of bacteriostasis and sterilization. But there are not any reports about whether abusage of bitter cold traditional Chinese medicines can cause the similar katogene as antibiotics. This study was undertaken to try to investigate the influence of Coptidis Decoction for Detoxification on the mice in the aspects of the changes of intestinal flora,the intestinal tissue damages and so on.Look forward to provide reference data for the clinical rational use of heat-cleaning and detoxifying traditional Chinese medicine.Method1.Constract the blank group,imbue with the saline water. The mice in blank group were given gastric lavage dally for 14 days by using 0.6ml saline water.2.Constract the control groups of enteric dysbacteriosis mice,use the lincomycinThe mice in study groups were given gastric lavage daily for 14 days by using 60mg lincomycin hydrochloride.3.Constract the study groups,including the higher dose group and the lower dose group,imbue with different doses of Coptidis Decoction for Detoxification.The mice in higher dose group were given gastric lavage daily for 14 days by using 0.15g Coptidis Decoction for Detoxification.The mice in lower dose group were given gastric lavage daily for 14 days by using 0.075g Coptidis Decoction for Detoxification.4.Observe the appearance,behavior,diet and so on of the mice in each groups.5.Examine the amount of Bacillus bifidus,Bacillus acidi lactici,enterococci and enteric bacilli in the appendix content of each mice,flowing the procedures of weighing,clariflxation,diluting,dripping,spreading, culturing and counting,investigate the effects of Coptidis Decoction for Detoxiflcation to the intestinal track from the changing of intestinal flora.6.Grind the spleen and mesenteric lymph nodes separately,culture and examine if there were bacteria translocation.7.Weigh the appendixes of each mice,and calculate the cecal index.Make the pathological section of ileum,analysis the affections of Coptidis Decoction for Detoxification to the intestinal track from the degree of intestinal tissue injury.8.Use the ANOVA modular of software package SPSS 13.0 as the statistical method to support data analysis,use the student-t test for comparing two means based on two groups of data under completely randomized design.Results1.Mice in the blank group eat well,move actively,clothing hair is smooth and even,have the normal faces,and the body weight gain.The intestinal flora keep in a normal balance,the ileal mucous membrane and the appendix are both in normal status,without edema.2.Mice in the control groups ingest fewer food,depressed and slinking, slow-moving.The clothing hair is dim,and the mice are algid,crowded, passage of loose stools,and lose weight.About 50%of the mice have the bacterial translocation from intestinal track to the mesenteric lymph nodes. The ratio of enterococcis to enteric bacilli of intestinal flora is larger than 1 to 3,Bacillus bifidus,Bacillus acidi lactici,enterococci and enteric bacilli are all different from the blank groups.They suffered from the dysbacteria,and desquamation of ileal mucous membrane and obviously edema of the appendix.3.Mice in the higher dose group ingest fewer food,depressed and slinking, slow-moving.The clothing hair is dim,feces are wispy and wet,and weight is losing.About 20%of the mice suffered from the bacterial translocation from intestinal track to the mesenteric lymph nodes.The ratio of enterococcis to enteric bacilli of intestinal flora is larger than 1 to 3,Bacillus bifidus, Bacillus acidi lactici,enterococci and enteric bacilli are all different from the blank groups.They suffered from the dysbacteria,and desquamation of ileal mucous membrane and obviously edema of the appendix.4.Mice in the lower dose group eat well,move actively,clothing hair is smooth and even,have the normal faces,and the body weight gain.The intestinal flora keep in a normal balance,the ileal mucous membrane and the appendix are both in normal status.ConclusionThe mice in the control group,which use the antibiotics,suffered from the dysbacteria of intestinal flora,and the similar symptoms emerged from the mice of higher dose group with gastric lavage of Coptidis Decoction for Detoxification.This situation suggested that,large dosages and/or long term usage of Coptidis Decoction for Detoxification have the similar injury effect as antibiotics.The Coptidis Decoction for Detoxification can reduce the amount of enterococci under the lower dose usage,and maybe have the influence of regulating the balance of intestinal flora.
