Telomerase, a tumor marker, is specifically expressed in cancer cells. In physiological conditions, most normal cells do not express telomerase, wherese 90 percent of tumor cells show a high level expression of telomerase. The core component of telomerase-human telomerase reverse transcriptase (hTERT) is a key determinant of telomerase activity.Bax gene is the main pro-apoptotic gene of Bcl-2 gene family, and Bax and bcl-2 regulate apoptosis as homologous or heterologous dimer. With special activity of hTERT promoter in tumor cells, CMV-pct-bax was constructed. The Bax expression is reversly associated with the expression of Bcl-2 in MCF-7. MTT experiments showed that cell growth was inhibited by Bax expression and Bax expression also induced apoptosis in MCF-7cells. In conclusion, CMV-pct-bax may be a novel and promising tool for targeting cancer therapy.
