| Stem cell which contains adult stem cell(ASC), embryonic stem cell(ESC), induced potential stem cell (IPS) has multi-differentiation ability ,so it plays a important role in the cell therapy and tissue engineering. ASC is superior to ESC, because it is without ethic, immunogenic problems and it is easy to isolate and culture and use in the clinic. Bone marrow mesenchymal stem cell(BMSC) and adipose derived mesenchymal stem cell(ADSC) have been proved to contribute to the cardiac muscle, skin and bone injuries repair and so on. But aspiration of BMSC and ADSC bring great pain and risk of infection to the patients. And also they curb the stem cell as a"drug"in the future clinical application, because few people want to donate bone marrow. People pay more attention to the blood stem cell ,because it has more superiorities, such as aspiration safety ,little pain to patients and general resource(blood back). Hemopoietic stem cell (HSC) has been proved to reconstruct immune system and cure ischemic disease of lower extremity , myocardial damage repair and so on. Whether PBMSC is in the peripheral blood or not without mobilization , it is on the debate. It is reported that PBMSC can contribute to bone injury repair and neuronal injury repair . But whether PBMSC can contribute to wound healing ,it is not repored. Our experiments is to improve the efficiency of isolation and culture of PBMSC firstly and test the function of PBMSC in the wound healing in the later.(1) Under the condition of wound back of rats, the number of PBMSC has been improved.(2) The supernatant BMSC contains many kinds of factors ,which can improve the efficiency of isolation and culture of PBMSC.(3) PBMSC has the property of mesenchymal stem cell(MSC) , which is negative for CD34 , CD45 and can differentiate to adipocyte and osteocyte.(4) PBMSC can contribute to wound healing similar with BMSC and be used for the seed cells of cell therapy and tissue engineering skin.
