Objective:This study was designed to investigate the effects of mesenchymal stem cells(MSCs)on remolding of heart after myocardium infarction in rats.Methods: MSCs from SD rats were cultured in serum-free medium or serum-free medium under normoxic or anoxic conditions for 24 hours and then the conditioned medium were collected.Cardiac fibroblasts(CFs)were cultured in the above MSCs medium.The gelatinolytic activity of MMP in culture medium of CFs was detected by Gelatin Zymography.Immunoblot was applied to determine the effect of hypoxic preconditioning on ERK/p-ERk pathway.Result:The synthesis and activity of MMPs secreted by CFs were significantly lower in AP-MSCs medium group than in the MSCs medium group and control group.Ratio of p-ERK/ERK in AP-MSCs medium group assayed by western-blotting was increased.Conclusion:The result indicated that anoxia-preconditioning enhanced the capacity of MSCs to reduce the ventricular remolding after MI partly by interfering with MMPs secretion of cardiac fibroblasts.
