Objective:To investigate the influence of cytokines correlating to Th1/Th2 cells on immune effect of hepatitis B vaccination,analyze the invalidation mechanism of hepatitis B vaccination for providing the evidence to elevate the achievement ratio of vaccination and prevent the hepatitis B infection effectively.Methods:901 college students aged 21 to 24 year old who had received standard courses of vaccination were select to be the study subjects.According to the different immune effect,the subjects were divided into two groups:the non-or-hypor-responders of anti-HBs negative and the normal or strong responders of anti-HBs positive;The concentration of IFN-γ,IL-4 cytokines correlating to Th1/Th2 in the subject serum were detected by the double antibody sandwich enzyme linked immununosorbent assay(ELISA).Results:①The IFN-γmean concentration of the non-or-hypor-responder group and the normal or strong responder group was 3.88±4.57 pg/ml and 7.49±7.05 pg/ml respectively,the IFN-γmean concentration of the non-or-hyporresponders is much less than that of the normal or strong responder to hepatitis B vaccination(P<0.01);②The IL-4 mean concentration of the non-or-hyporresponder groups and the normal or strong responder groups was 2.42±1.64 pg/ml and 5.25±3.08 pg/ml respectively,the IL-4 mean concentration of the non-or-hypor-responders is also much less than that of the normal or strong responder to hepatitis B vaccination(P<0.01).Conclusions:The concentration of IFN-γand IL-4 in the non-or-hyporresponders is significantly less than that of the normal or strong responder to hepatitis B vaccination.The study results show that the levels of cytokines IFN-γand IL-4 correlating to Th1/Th2 cells may affect on the immune effect of hepatitis B vaccination,and suggest that IFN-γand IL-4 could be used as the adjuvant in vaccination of hepatitis B to achieve success of vaccination.
