Objective:To detect the expression of CD151 mRNA and protein and the expression of integrinα3,β1 protein,then investigate the relationship with the two in thyroid neoplasms tumorgenesis.Methods:1.we detected CD151 mRNA in 44 cases of fresh thyroid surgical specimen(26 cases of thyroid papillary carcinoma,10 cases of follicular adenoma and 8 cases of away from tumor 2cm normal thyroid gland tissue) using RT-PCR,and analyzed with clinical data.2.we detected the expression of CD151 and integrinα3,β1 protein in 86 case of thyroid specimen(including the 44 cases in RT-PCR,in all,64 cases of thyroid papillary carcinoma,12 cases of follicular adenoma and 10 cases of away from tumor 2cm normal thyroid gland tissue)using immunohistochemistry Envision two stage method,and analyzed with clinical data.Result:1.The expression of CD151 mRNA in thyroid papillary carcinoma is 2.13±0.91,in follicular adenoma and normal thyroid tissue is 1.44±0.15 and 1.09±0.16;The different between carcinoma with adenoma and normal tissue both have statistical significance(P<0.01),and the different between adenoma with normal tissue also has statistical significance(P<0.05);There is no association between the expression of CD151 mRNA in the age,gender,tumor size,and lymph nodes metastasis of the thyroid papillary carcinoma cases.2.The result of detecting the expression of CD151 protein in the same specimen as which detected mRNA using RT-PCR,is coincidence with the expression of mRNA.the positive rate of carcinoma,adenoma and normal tissue is 88.5%(23/26),50%(5/10)and 0%(0/8).There is statistical significance in group comparison(P<0.01),but there is no association between the expression in the age,gender,tumor size and lymph nodes metastasis(P>0.05).A positive correlation was observed between CD151 mRNA and protein expression in thyroid tissue(r=0.511,P<0.01).3.Detected the 86 thyroid tissue,we found that CD151 positive rate is 87.5%(56/64)in carcinoma,41.7%(5/12)in follicular adenoma and 0%(0/10)in para-tumor normal thyroid tissue.The expression in carcinoma is obviously higher than other two groups,the different is statistical significance(P<0.01). No association was observed between the expression in the age,gender,tumor size and lymph nodes metastasis.The expression of integrinα3 in thyroid papillary carcinoma,follicular adenoma and normal thyroid tissue is 78.1%(50/64),33.3%(4/12)and 0%(0/10),the expression in carcinoma is higher than other groups(P<0.01),but there is no association between the expression and age,gender,tumor size and lymph nodes metastasis in thyroid carcinoma cases(P>0.05),the expression of integrinβ1 in thyroid papillary carcinoma, follicular adenoma and normal thyroid tissue is 71.9%(46/64),25%(3/12)and 0%(0/10).The expression in carcinoma is higher than other groups(P<0.01), but there is no association between the expression and age,gender,tumor size and lymph nodes metastasis in thyroid papillary carcinoma cases(P>0.05). 4.A positive correlation was observed between CD 151 and integrinα3,β1 in thyroid tissue(r=0.565,0.363,0.504,P<0.01). Conclusion:1.The expression of CD151 in normal thyroid tissue,follicular adenoma and thyroid papillary carcinoma is progressively increasing,prompted that CD151 high expression may play a promotive effect on the tumorigenesis of thyroid papillary carcinoma.2.The expression of integrinα3 andβ1 subunit in thyroid papillary carcinoma is obviously higher than follicular adenoma and normal thyroid tissue, suggested that the expression was relevant to thyroid papillary carcinoma.3.Since the expression of CD151 and integrinα3,β1 in thyroid papillary carcinoma is positive correlating,suggected that the two maybe play an effect on thyroid papillary carcinogenesis,the detection of them in common can provide reference of diagnosing thyroid papillary carcinoma.
